摘　　要：目的:探讨永离有丝分裂基因A相关激酶6(never in mitosis gene A-related expressed kinase 6,NEK6)在卵巢癌组织中的表达水平,分析其表达与临床病理因素及预后之间的关系。方法:收集临床上卵巢癌患者的肿瘤及正常卵巢组织,用实时定量PCR(real-time quantitative PCR,RT-qPCR)和蛋白质印迹(Western Blot)检测卵巢癌组织和正常卵巢组织中NEK6的表达情况,并用免疫组织化学的方法检测110例卵巢癌患者中NEK6的表达水平,分析其表达水平与年龄、病理类型、肿瘤分期、组织分级、转移之间的关系,用Kaplan-Meier生存分析法分析NEK6在卵巢癌中的表达水平与患者生存期之间的关系。结果:RT-qPCR、Western Blot及免疫组织化学结果均显示卵巢癌组织中的NEK6表达水平显著高于正常卵巢组织(均P<0.01);NEK6在卵巢癌患者肿瘤组织中的表达水平与卵巢癌的组织分级(P=0.008)、转移(P=0.006)呈正相关;Kaplan-Meier分析显示NEK6高表达水平的患者总生存期显著低于NEK6低表达患者(P<0.01)。结论:NEK6在卵巢癌患者的肿瘤组织中高表达,并影响卵巢癌的转移,提示NEK6可能参与卵巢的发生发展,可以作为潜在的卵巢癌预后指标。Objective:To detect the expression of never in mitosis gene A-related expressed kinase 6(NEK6)in ovarian cancer tissue,and to investigate the correlation between its expression and clinical-pathological parameters as well as prognosis.Methods:Collecting tumor and normal tissues from clinical ovarian cancer patients,the real-time quantitative PCR(RT-qPCR)and Western Blot assays were used to detect the expression of NEK6 in ovarian cancer and normal ovarian tissues,and immunohistochemistry was used to detect the expression of NEK6 in 110 ovarian cancer patients.The relationship analysis was performed between its expression and age,pathological type,tumor stage,tissue grading,and metastasis.Kaplan-Meier survival curve was used to analyze the relationship between NEK6 expression in ovarian cancer and patient survival.Results:RT-qPCR,Western Blot,and immunohistochemistry results all showed that NEK6 levels in the tissues of ovarian cancer were significant higher than those in normal ovarian tissues(all P<0.01).The expression level of NEK6 was positively correlated with the tissue grading(P=0.008)and metastasis(P=0.006)of ovarian cancer.Kaplan-Meier analysis showed that patients with high expression of NEK6 had significantly lower overall survival than those with low expression(all P<0.01).Conclusion:NEK6 was highly expressed in the tumor tissue of ovarian cancer patients and affects the metastasis of ovarian cancer,suggesting that NEK6 may be involved in the occurrence and development of the ovary and can serve as a potential prognostic indicator for ovarian cancer.
