摘  要:选取化妆品常用的22种活性原料,分别利用紫外分光光度计法和SDS-PAGE法进行体外抑制木瓜蛋白酶活性实验;利用Franz扩散池和人体功效测试检验纤连蛋白透皮过程与抗皱功效,间接证明对木瓜蛋白酶的抑制效果。结果表明,金缕梅提取物抑制酶解反应获得的纤连蛋白浓度最高,其他依次为无患子提取物、海茴香提取物、燕麦提取物、高山火绒草提取物、卡瓦胡椒提取物,对纤连蛋白的透皮与改善眼角皱纹有显著性差异。Papain,which is a typical representative of Cysteine protease,was selected for screening natural inhibitor.Twenty-two commonly used active ingredients in cosmetics were chosen for in vitro papain inhibition experiments.The optical density(OD)of the reaction end product was measured using UV spectrophotometry and SDS-PAGE to assess the inhibitory effects of ten natural active ingredients.The Franz diffusion cell and human efficacy test were used to investigate the inhibition effect of fibronectin in the transdermal process and anti-wrinkle effect,respectively.The results showed that Hamamelis virginiana extract exhibited the highest concentration of fibronectin(FN1)in the enzymatic hydrolysis reaction,followed by Sapindus Saponaria extract,Crithmum maritimum extract,Avena sativa extract,Leontopodium brachyactis extract.Hamamelis virginiana extract and FN1 significantly improved the depth,length,and area of wrinkles at the corners of the eyes.
