摘  要:目的基于亚氨基二丙腈(IDPN)动物模型的实验文献,阐述IDPN模型的效度研究现状,为抽动障碍(TD)实验研究合理选择动物模型提供参考。方法检索2013~2023年中英文数据库,选择国内学者以IDPN作为诱导剂的动物实验文献进行描述性分析。结果共纳入140篇文献,分析提示近10年国内学者的年发文量略有下降,多巴胺、纹状体、大鼠为高频关键词。研究者常使用3~4周龄的SD雄性大鼠,多采用150 mg/(kg·d)、250~300 mg/(kg·d)的IDPN腹腔注射1周建立TD模型,Diamond刻板行为评分≥1分标志造模成功。模型鼠头颈部刻板运动可持续8周,脑组织及血液中的神经递质和细胞因子含量是常用的模型结构效度评价指标,多数文献显示模型鼠纹状体多巴胺D1受体(DRD1)、多巴胺D_(2)受体(DRD_(2))、5-羟色胺(5-HT)、谷氨酸(GLU)及血多巴胺(DA)含量升高,纹状体DA、5-HT转运体(SERT)含量降低,脑组织和血白细胞介素(IL)-1及干扰素-γ(IFN-γ)含量升高。脑组织出现一定程度结构改变。结论IDPN模型具有稳定持续的表面效度,涉及皮质-纹状体-丘脑-皮质(CSTC)环路神经递质失衡的结构效度,以及拮抗多巴胺D_(2)受体的预测效度。Objective To explore the research status of the validity of iminodipropionitrile(IDPN)animal model based on the related experimental literature,and to provide reference for the rational selection of animal model in the experimental study of Tic disorder(TD).Methods The Chinese and English databases from 2013 to 2023 were searched,and the animal experimental literature with IDPN as inducer by domestic scholars was selected for descriptive analysis.Results A total of 140 articles were included,and the analysis showed that the number of articles published by domestic scholars decreased slightly in recent 10 years,and dopamine,striatum and rat were the high-frequency keywords.Researchers often used SD male rats of 3~4 weeks old,and usually used IDPN[150 mg/(kg·d),250~300 mg/(kg·d)]for one week to establish TD model.The Diamond stereotyped behavior score≥1 indicated that the model was successful.The stereotyped movement of the head and neck of model rats could last for 8 weeks,and the contents of neurotransmitters and cytokines in brain tissue and blood were commonly used to evaluate the structural validity of the model.Most literature showed that the contents of DRD1,DRD_(2),5-HT,GLU in striatum and DA in blood of the model rats were increased,and the contents of DA and SERT in the striatum were decreased,while the contents of IL-1 and IFN-γin brain tissue and blood were increased.There were some structural changes in brain tissue.Conclusion The IDPN model has a stable and sustained face validity,which involves the structural validity of neurotransmitter imbalance in CSTC loop and the predictive validity of antagonizing DRD_(2).
