摘 要:水苏糖作为一种天然益生元,可促进乳酸菌增殖,但水苏糖在乳酸菌中的代谢途径仍未被完全解析。该研究尝试建立一种检测水苏糖及其代谢产物的高效液相色谱-蒸发光散射(high performance liquid chromatography-evaporative light-scattering detector, HPLC-ELSD)方法,通过水苏糖及其代谢产物含量的动态变化为阐释水苏糖在乳酸菌中的代谢途径提供技术支撑。结果表明,使用Waters BEH Amide色谱柱结合ELSD检测器,洗脱条件为V(乙腈)∶V(水)=80∶20、流动相流速为1.0 mL/min, ELSD检测器条件为温度60℃,载气流速1.0 mL/min时,水苏糖及其代谢产物能实现较好分离,且检测重现性好,稳定性高,说明该方法适用于水苏糖及其代谢产物含量的检测。Stachyose,as a natural prebiotic,can promote the proliferation of lactic acid bacteria,but the metabolic pathway of stachyose in lactic acid bacteria is still completely resolved.This study attempts to establish a high-performance liquid chromatography-evaporative light-scattering detector(HPLC-ELSD)method for the detection of stachyose and its metabolites.The dynamic changes of stachyose and its metabolites were intended to provide technical support for the interpretation of the metabolic pathway of stachyose in lactic acid bacteria.Results showed that using Waters BEH Amide chromatographic column combined with ELSD detector,the elution conditions were acetonitrile(80%)∶water(20%),the flow rate of mobile phase was 1.0 mL/min,the temperature of ELSD detector was 60℃,and the flow rate of carrier gas was 1.0 mL/min.Stachyose and its metabolites can be well separated,and the detection reproducibility is good and the stability is high,indicating that this method can be used for the detection of stachyose and its metabolites.
