摘　　要：目的探讨长链非编码RNA肿瘤易感性候选基因9(lncRNA CASC9)、YKT6在口腔鳞状细胞癌(OSCC)中表达意义及预后价值。方法选取2016年1月至2018年1月在陕西中医药大学附属医院诊治的110例OSCC患者作为研究对象。采用实时荧光定量PCR法检测患者OSCC癌组织及癌旁组织lncRNA CASC9、YKT6 mRNA表达。采用免疫组化检测OSCC癌组织及癌旁组织YKT6蛋白表达。Kaplan-Meier曲线分析不同lncRNA CASC9、YKT6 mRNA表达对OSCC患者预后的影响。Cox回归模型分析影响OSCC预后的因素。结果OSCC癌组织lncRNA CASC9、YKT6 mRNA表达分别为3.12±0.57、2.69±0.42,均明显高于癌旁组织(依次为1.02±0.25、1.13±0.21),差异有统计学意义(t=35.386、34.843,P<0.05)。OSCC癌组织YKT6蛋白阳性率为81.82%(90/110),高于癌旁组织[9.09%(10/110)],差异有统计学意义(χ^(2)=117.333,P<0.05)。OSCC癌组织lncRNA CASC9与YKT6 mRNA表达呈正相关(r=0.788,P<0.001)。TNM分期Ⅲ~Ⅳ期、低分化及有淋巴结转移的OSCC癌组织lncRNA CASC9、YKT6 mRNA表达高于TNM分期Ⅰ~Ⅱ期、高中分化及无淋巴结转移的OSCC癌组织lncRNA CASC9、YKT6 mRNA表达,差异有统计学意义(均P<0.05)。lncRNA CASC9高表达组、YKT6 mRNA高表达组5年累积生存率低于lncRNA CASC9低表达组、YKT6 mRNA低表达组,差异有统计学意义(Log-rankχ^(2)=7.080、8.741,P=0.008、0.003)。lncRNA CASC9高表达、YKT6 mRNA高表达、TNM分期Ⅲ~Ⅳ期、低分化及有淋巴结转移是OSCC患者预后危险因素。结论OSCC癌组织中lncRNA CASC9、YKT6表达升高,两者可作为评估OSCC患者预后的肿瘤标志物。Objective To investigate the expression significance and prognostic value of long non-coding RNA tumor susceptibility candidate gene 9(lncRNA CASC9)and YKT6 in oral squamous cell carcinoma(OSCC).Methods A total of 110 patients with OSCC treated in the Affiliated Hospital of Shaanxi University of Traditional Chinese Medicine from January 2016 to January 2018 were selected as the study objects.The mRNA expression of lncRNA CASC9 and YKT6 in OSCC cancer tissue and adjacent tissues was detected by real-time fluorescence quantitative PCR.Immunohistochemistry was used to detect YKT6 protein expression in OSCC cancer tissue and adjacent tissues.Kaplan-Meier curves were used to analyze the influence of different lncRNA CASC9 and YKT6 mRNA expression on the prognosis of OSCC patients.Cox regression model was used to analyze the prognostic factors of OSCC.Results The lncRNA CASC9 and YKT6 mRNA expression in OSCC cancer tissues were 3.12±0.57 and 2.69±0.42,respectively,which were significantly higher than those in adjacent tissues(1.02±0.25,1.13±0.21),and the differences were statistically significant(t=35.386,34.843,P<0.05).The positive rate of YKT6 protein in OSCC cancer tissues was 81.82%(90/110),which was higher than that in adjacent tissues[9.09%(10/110)],and the difference was statistically significant(χ^(2)=117.333,P<0.05).There was a positive correlation between lncRNA CASC9 mRNA expression and YKT6 mRNA expression in OSCC cancer tissues(r=0.788,P<0.001).The lncRNA CASC9 and YKT6 mRNA expressions in cancer tissues of OSCC with TNM stagesⅢtoⅣ,low differentiation and lymph node metastasis were higher than those in cancer tissues of OSCC with TNM stagesⅠtoⅡ,high school differentiation and no lymph node metastasis,the difference was statistically significant(all P<0.05).The 5-year cumulative survival rate of lncRNA CASC9 high expression group and YKT6 mRNA high expression group was lower than that of lncRNA CASC9 low expression group and YKT6 mRNA low expression group,and the difference was statistically si
