摘 要:白凤菜,是一味闽南地区及中国台湾著名的中草药,其作用多样,但近年来研究较少,特别是分子研究匮乏。本研究选择Illumina HiSeq~(TM)平台对白凤菜的叶和茎进行了高通量测序,研究结果表明:一共拼接成93 975个高质量的unigene,其N50为1 146 bp,平均长度为624.78 bp;与基因本体数据库等七大数据库进行实验结果对比辨析,共有56 455个unigene至少顺利注释一个数据库,全部注释的有2 005个unigene。在白凤菜叶对根差异表达中共获得3 286个unigene,其中上升趋势表达1 852个,下降趋势表达1 434个。利用KEGG数据库,找到参与类黄酮生物合成的有39个unigene,具有显著差异的有11个,分布在12个合成酶中。此结果为进一步研究白凤菜黄酮类化合物生物合成和调控的分子机制提供理论基础。Gynura formosana Kitam.is a famous Chinese herbal medicine in southern Fujian and Taiwan,China.Its functions are diverse,but there is little research in recent years,especially molecular research.This study selected Illumina HiSeqTM the platform carried out high-throughput sequencing on the leaves and stems of G.formosana,and the research clarified that a total of 93975 high-quality unigenes were spliced,with an N50 of 1146 bp and an average length of 624.78 bp;compared with seven major databases such as GO database,56455 unigenes annotated at least one database successfully,and 2005 unigenes annotated all.A total of 3286 unigenes were obtained in the differential expression of roots in the leaves of G.formosana,of which 1852 were expressed with an upward trend and 1434 were expressed with a downward trend.Using the KEGG database,39 unigenes were found to be involved in flavonoid biosynthesis,with 11 showing significant differences,distributed among 12 synthetases.These results provide a theoretical basis for further research on the molecular mechanism of biosynthesis and regulation of flavonoids in G.formosana.
