摘　　要：目的探讨胃癌患者外周血及组织中高迁移率族蛋白A1(high mobility group AT-hook1,HMGA1)表达及临床意义。方法应用酶联免疫吸附实验及免疫组织化学染色胃癌患者病理石腊包埋组织进行外周血及组织中HMGA1的表达检测;检索数据库分析HMGA1在胃癌肿瘤组织的表达以及总生存期;采用Western Blot及qRT-PCR检测胃癌肿瘤组织及癌旁组织中HMGA1蛋白及mRNA的表达。结果胃癌患者外周血中HMGA1的表达高于健康体检者,差异有统计学意义(P<0.001);在胃癌患者外周血中HMGA1的表达在患者的年龄、分化程度、Lauren分型、浸润程度及有无淋巴结转移等方面,差异具有统计学意义(P<0.001);胃癌患者行根治手术后第1天、第3天、第5天、第7天的HMGA1的表达高于胃癌患者手术前,差异有统计学意义(P<0.001);高表达HMGA1与胃癌的生存期有关,且肿瘤组织中HMGA1蛋白及mRNA的表达水平增加(P<0.05)。结论检测胃癌患者外周血和肿瘤组织中HMGA1的表达对临床的诊断和治疗有一定的参考价值。Objective To investigate the expression and clinical significance of high mobility group AT-hook 1(HMGA1)in peripheral blood and tissues of gastric cancer patients.Methods Enzyme-linked immunosorbent assay and immunohistochemical staining were used to detect the expression of HMGA1 in peripheral blood and paraffin-embedded tissues of gastric cancer patients.Database analysis was searched to analyze the expression of HMGA1 in gastric cancer tissues and its relation to overall survival.Western Blot and qRT-PCR were used to detect the expression of HMGA1 protein and mRNA in tumor and adjacent tissues of gastric cancer.Results HMGA1 expression in the peripheral blood of gastric cancer patients was higher than that of healthy medical examiners,with a statistically significant difference(P<0.001),and the difference was statistically significant with age of the patients,degree of differentiation,Lauren typing,degree of infiltration,and whether or not there were lymph node metastases(P<0.001).HMGA1 expression was significantly higher on days 1,3,5,and 7 post-radical surgery compared to pre-surgery levels,and the difference was statistically significant(P<0.001).High expression of HMGA1 was associated with overall survival(OS)of gastric cancer,and increased HMGA1 protein and mRNA expression levels were observed in tumor tissues(P<0.05).Conclusion There is certain valuable reference in HMGA1 expression detection from peripheral blood and tumor tissues of gastric cancer patients for clinical diagnosis and treatment.
