摘  要:建立了高效液相色谱(HPLC)法测定大麦苗及其产品中2种黄酮大麦黄苷和皂草黄苷的含量,研究了10个品种的青稞苗、 2个品种的皮大麦苗、 1个未知品种的大麦苗以及11个大麦苗产品中的大麦黄苷和皂草黄苷含量的差异。样品采用60%(V/V)乙醇作为提取溶剂,液料比为50 mL/g,室温超声30 min,采用C18(250 mm×4.6 mm, 5μm)色谱柱分离,甲醇/0.5%甲酸水溶液(30/70, V/V)为流动相进行洗脱,对大麦黄苷在355 nm、皂草黄苷在338 nm波长下检测。在优化条件下,大麦黄苷和皂草黄苷在0.200~100μg/mL范围内线性关系良好,加标回收率为96.2%~100%,大麦黄苷的检出限和定量限分别为9 mg/kg和29 mg/kg,皂草黄苷的检出限和定量限分别为12 mg/kg和39 mg/kg。12个已知品种中,除了青稞昆仑15号,皮大麦苗Golden promise和Bonus的2种黄酮含量普遍高于其它9个青稞品种;10个青稞品种中,昆仑15号的2种黄酮含量最高。11个大麦苗产品根据2种黄酮含量可分为6组,各组之间差异显著。大麦黄苷和皂草黄苷在14种非麦苗产品中均未检出,可作为区分它们的质量标志物。本方法适用于高黄酮含量大麦苗品种筛选以及大麦苗产品的质量控制。A high performance liquid chromatography(HPLC)method was established for the determination of barley seedlings and their products,and the differences in barley seedlings of 10 hulless barleys,two lemma barleys,an unknown variety,and 11 products of barley seedlings were studied.The samples were extracted with 60%(V/V)ethanol,and ultrasonicated for 30 min at room temperature with a liquid-solid ratio of 50 mL/g.The targets were separated on a C18(250 mm×4.6 mm,5μm)column and eluted with a mobile phase consisting of methanol/0.5%formic acid in water(30/70,V/V).The detection wavelengths for lutonarin and saponarin were 355 and 338 nm,respectively.Under the optimum conditions,the two compounds of lutonarin and saponarin showed good linear relationships in the range of 0.200-100μg/mL.The spiked recoveries ranged from 96.2%to 100%.The limit of detection(LOD)and limit of quantitation(LOQ)for lutonarin were 9 mg/kg and 29 mg/kg,respectively,while the LOD and LOQ for sapnoarin were 12 mg/kg and 39 mg/kg,respectively.Results indicated that in twelve known varieties,except the hulless barley of Kunlun 15,the two flavones in the lemma barleys of Golden promise and Bonus were higher than that in the other 9 hulless barleys,and the two flavones in Kunlun 15 were the highest among the 10 hulless barleys.The 11 products of barley seedlings could be divided into 6 groups,depending upon the contents of the two flavones,which exhibited significant differences.On the contrary,the two flavones were not detected in 14 non-barley seedling products,which can be used as quality markers to distinguish them from barley seedling products.The method is suitable for screening barley seedlings with high flavonoid content,and is applicable to the quality control of products of barley seedlings.
