基于抗氧化活性响应曲面法优化桔梗汤中桔梗-甘草配伍

摘  要:采用响应曲面法对桔梗汤中桔梗-甘草最佳配伍比例及其提取工艺进行优化。采用DPPH法比较不同配伍比例桔梗汤提取物抗氧化能力。以料液比、提取时间和提取次数为自变量,以单因素试验结果为基础,以甘草苷、党参炔苷、桔梗皂苷D、甘草酸总含量为响应值,采用Box-Behnken响应面法优化最佳比例的提取工艺。结果表明:桔梗汤提取物对DPPH有较好的清除效果,其中1∶2比例对DPPH清除能力最强,其IC_(50)值0.444 mg/mL。响应面结果显示,以料液比1∶22 (g∶mL),提取时间1.3 h,提取3次时上述成分含量最高。通过体外抗氧化能力探究桔梗汤最佳配伍比例,并优化其提取工艺,为桔梗汤制剂的深入开发及其临床应用提供理论支撑。Response surface methodology(RSM)was used to study the optimal compatibility ratio and extraction process of Radix Platycodi-Radix et Rhizoma Glycyrrhizae in Jie-geng decoction(JGD).DPPH was used to compare the antioxidant capacity of extracts from different compatibility ra⁃tios of JGD.With solid-liquid ratio,extraction time and extraction frequency as independent vari⁃ables,based on the results of single-factor experiment,and with the total contents of glycyrrhizin,lo⁃betyolin,platycodin D and glycyrrhizin as response values,Box-Behnken RSM was adopted to opti⁃mize the extraction process with the optimal proportion.The results showed that JGD extracts had better scavenging effect on DPPH,among which 1∶2 ratio had the strongest scavenging effect,with an IC_(50) value of 0.444 mg/mL.The response surface results showed that when solid-liquid ratio was 1∶22(g∶mL),extraction time was 1.3 h,and extraction was performed three times,the content of the above index components was the highest.In conclusion,through in vitro antioxidant capacity,the optimal compatibility ratio of JGD has been explored and the extraction process has been optimized, so as to provide theoretical support for further development and clinical application of it.
