摘 要:目的 研究CPNE1在小细胞肺癌转移中的作用及其机制。方法 通过GEO数据库分析CPNE1在小细胞肺癌患者与正常人组织中的表达水平。通过cBioportal数据库分析小细胞肺癌组织中CPNE1与AXL、MMP2、MMP9及SNAI1表达的相关性。利用qRT-PCR和Western Blot检测CPNE1在小细胞肺癌细胞株内表达情况以及干扰CPNE1对AXL及上皮间充质转移(EMT)信号通路mRNA和蛋白的影响。利用Transwell实验检测细胞迁移和侵袭的变化。结果 GEO数据库提示CPNE1在小细胞肺癌组织中表达高于正常组织(P<0.05)。此外,与Beas-2B相比,CPNE1在小细胞H446和H196细胞株中高表达。敲低CPNE1后H446细胞的迁移和侵袭能力下降(P<0.05)。生物信息学分析显示,CPNE1与AXL以及EMT相关标记物MMP2、MMP9及SNAI1表达均呈正相关(P<0.05)。同时敲低CPNE1后AXL及MMP2、MMP9、SNAI1等表达均下调(P<0.05)。结论 CPNE1在小细胞肺癌中高表达,并可通过影响AXL介导的EMT通路调控细胞转移。Objective To investigate the role and mechanism of CPNE1 in metastasis of small cell lung cancer.Methods The expression level of CPNE1 in small-cell lung cancer patients and normal subjects was analyzed by the GEO database.The correlation between CPNE1 expression and AXL,MMP2,MMP9,and SNAI1 expression in small lung cancer tissues was analyzed by the cBioportal database.qRT-PCR and Western Blot were used to detect the expression of CPNE1 in small cell lung cancer cell lines and the effect of interference with CPNE1 on mRNA and protein of AXL and epithelial-mesenchymal transfer(EMT)signaling pathway.The changes in cell migration and invasion were detected by Transwell assay.Results GSE database showed that CPNE1 expression was elevated in SCLC tissues than in normal tissues(P<0.05).When compared with Beas-2B,CPNE1 was highly expressed in H446 and H196 cells.The migration and invasion ability of H446 cells decreased after CPNE1 knockdown(P<0.05).Bioinformatics analysis showed that CPNE1 was positively correlated with AXL and the expression of EMT-related markers MMP2,MMP9 and SNAI1(P<0.05).The expression of AXL,MMP2,MMP9 and SNAIL were down-regulated after CPNE1 was depressed(P<0.05).Conclusion CPNE1 is highly expressed in small-cell lung cancer and regulates cell metastasis by affecting the AXL-mediated EMT pathway.
