响应面法优化构树叶多糖酶法提取工艺的研究

摘  要:利用纤维素酶法提取构树叶多糖,考查了料液比、提取温度、提取时间和纤维素酶添加量4个单因素对多糖得率的影响。在此基础上,利用响应面法优化构树叶多糖的酶法提取工艺。响应面得到的最佳工艺条件为料液比1∶30(g∶mL),提取温度58℃,提取时间160 min,纤维素酶添加量0.95%,在此条件下,多糖提取率为17.25%,验证试验结果与模型预测值相符。Cellulase was investigated to extract polysaccharides from Broussonetia papyrifera leaf,the effects of solid/liquid rate,temperature,time and cellulase dosage on the yield of polysaccharides were studied.The optimal process conditions were obtained through response surface and solid/liquid rate was 1∶30(g∶mL),temperature was 58℃,time was 160 min and cellulase dosage was 0.95%,under this condition the polysaccharide yield was 17.25%,which verified that the test results were consistent with the predicted values of the model.
