摘 要:离体脊髓切片是医学研究生电生理科研实验中常用的活体组织。传统脊髓切片方法已沿用多年,由于实验技术条件所限,该方法仅适用于幼鼠。本实验室对传统实验方法进行了多项优化,包括加入心脏灌注手术,简化取髓剥膜步骤,改变溶液配方等措施。改进后的实验方法在一定程度上提高了神经元的活性,并在成年大鼠脊髓切片上成功地进行膜片钳记录。Spinal cord slices are commonly used live tissue in electrophysiological experimental for medical graduate postgraduates.The traditional spinal cord slicing method has been employedfor numerous years.Owing to the limitations of experimental technical conditions,this method is merely applicable to young rats.A multitude of optimizations have been carried out on the experimental method in the article,including the addition of heart perfusion surgery,the simplification of the steps of taking the spinal cord and stripping the membrane,as well as the alteration of the solution formula and other measures.The improvedexperimental method has enhanced the activity of neurons to a certain extent,and successfully carried out patch clamp recordings on the spinal cord slices of adult rats.
