摘　　要：目的:探讨连云港汉族人群的ADH1B rs1229984和ALDH2 rs671多态性与胃癌、食管癌相关性。方法:以随机数字表法选取2021年1~12月在连云港市第二人民医院就诊的50例胃癌患者和50例食管癌患者作为研究对象,同期以随机数字表法选取200名汉族健康献血者纳入健康组。检测ADH1B rs1229984和ALDH2 rs671 SNP的多态性,探究连云港汉族人群的ADH1B rs1229984和ALDH2 rs671多态性与胃癌、食管癌相关性。结果:3组研究对象的性别、年龄、BMI、吸烟史、饮酒史等一般资料比较差异均无统计学意义(P>0.05),具有可比性。健康人群rs1229984 AA、AG和GG基因型的频率分别为42.5%(85例)、53.5%(107例)和4%(8例)。rs671的基因型频率AA为1%(2例),AG为26.5%(53例),GG为72.5%(145例),符合Hardy-Weinberg平衡(P>0.05),有群体代表性。分析结果显示,胃癌组和食管癌组患者的ALDH2 rs671 AA、AG、GG分布频率与健康组比较,差异有统计学意义(P<0.05);食管癌患者ADH1B rs1229984的AA、AG、GG基因分布频率与健康组比较,差异有统计学意义(P<0.05)。结论:胃癌和食管癌的基因分布组成比例与健康人群有显著性差异,ADH1B rs1229984和ALDH2 rs671多态性与连云港汉族人群胃癌、食管癌存在一定的相关性。Objective:To explore the associations of ADH1B rs1229984 and ALDH2 rs671 polymorphisms with gastric and esophageal cancer risks.Methods:Using a random number table method,100 patients(50 with gastric cancer and 50 with esophageal cancer)treated at the Second People′s Hospital of Lianyungang from January to December 2021 were randomly enrolled,along with 200 healthy Han ethnic blood donors serving as healthy group.The polymorphisms of ADH1B rs1229984 and ALDH2 rs671 SNPs were detected,and their associations with gastric cancer and esophageal cancer were investigated in the Han population in Lianyungang area.Results:Comparison of the general clinical data,including gender,age,BMI,smoking history and drinking history among the three groups of subjects showed no significant differences(P>0.05).In healthy group,the frequencies of rs1229984 AA,AG,and GG genotypes were 42.5%(85 cases),53.5%(107 cases),and 4%(8 cases),respectively.For rs671,the genotype frequencies were 1%(2 cases)for AA,26.5%(53 cases)for AG,and 72.5%(145 cases)for GG.Both sets of genotype frequencies complied with the Hardy-Weinberg equilibrium(P>0.05),indicating their representativeness in the population.The analysis results showed that the distribution frequencies of ALDH2 rs671 AA,AG and GG in gastric and esophageal cancer group patients were statistically different from those in healthy group patients(P<0.05).Significant differences were observed in the distribution frequencies of the ADH1B rs1229984 AA,AG and GG genotype,between esophageal cancer patients and healthy controls(P<0.05).Conclusion:The genetic distribution patterns in gastric cancer and esophageal cancer groups differ significantly from those in the healthy control group.The polymorphisms of ADH1B rs1229984 and ALDH2 rs671 have a certain association with gastric cancer and esophageal cancer in the Han population living in Lianyungang area.
