摘 要:为多维度评价电子束辐照灭菌技术在白花蛇舌草中的应用前景,研究考察了电子束辐照灭菌对白花蛇舌草微生物水平、化学成分含量、指纹图谱、体外抗氧化活性及细胞存活率的影响。结果表明,电子束辐照灭菌对白花蛇舌草中微生物具有明显杀灭作用。吸收剂量2~10 kGy时,白花蛇舌草中芦丁与车叶草苷含量、3种抗氧化活性、细胞存活率较灭菌前均无显著差异(P>0.05),吸收剂量大于10 kGy时,抗氧化活性及十六烷酸甲酯含量降低,建议安全吸收剂量为4~10 kGy。灭菌后白花蛇舌草指纹图谱较对照指纹图谱的相似度均大于0.90。液质联用(LC-MS)确认了指纹图谱中14个共有峰结构,灰色关联度分析(GRA)显示14个共有峰与3种抗氧化活性的关联度(r)均大于0.5,其中与1,1-二苯基-2-三硝基苯肼自由基(DPPH·)反应率超过50%的峰为8、10、11号峰。本研究可为电子束辐照灭菌技术在白花蛇舌草生产中的应用提供科学依据。To evaluate the application of electron beam irradiation sterilization technology in Herba Hedyoti Diffusae from multiple dimensions,its effect on the microbial level,chemical composition content,fingerprint,antioxidant activity,and cell survival rate of the sample materials was investigated.The results showed that irradiation could greatly sterilize the microorganisms in Herba Hedyoti Diffusae.The content of rutin and luteolin,as well as the three antioxidant activities and cell survival rate were not significantly different from these before sterilization(P>0.05)at an irradiation dose of 2-10 kGy.When the irradiation dose was greater than 10 kGy,the antioxidant activity in vitro decreased,and the content of hexadecane methyl ester decreased.It is recommended to use a safe irradiation dose of 4-10 kGy.After sterilization,the similarity between the fingerprint of Hedyotis diffusa and the control fingerprint was greater than 0.90.Liquid chromatography-mass spectrometry(LC-MS)confirmed the 14 compounds in the fingerprint spectrum.Grey correlation analysis(GRA)showed that the correlation degree r between the 14 compounds and the three antioxidant activities was greater than 0.5.Among them,peaks 8,10,and 11 had a reaction rate of over 50%with DPPH·.This study provided some scientific basis for the application of the electron beam technology in the production of Herba Hedyoti Diffusae.
