摘 要:目的 探讨超声引导下兔VX2肝肿瘤建模的成功率,以及紫杉醇(PTX)联合无水乙醇注射治疗(AEI)兔VX2肝脏肿瘤的有效性。方法 超声引导下将VX2肿瘤细胞注入兔肝脏内,2周后利用超声造影观察肿瘤生长情况,将建模成功的兔子分为6组:假手术组(注入2 mL生理盐水),临床单纯无水乙醇(AE)治疗(按肿瘤体积确定AE用量,C-AEI组),2 mL,AE联合不同浓度PTX混合液治疗[PTX依次为0 mg(AEI组)、12.5 mg(低剂量PTX组)、25.0 mg(中剂量PTX组)、37.5 mg(高剂量PTX组)]。1周之后行超声造影评估肿瘤坏死率(RNR),处死动物后获取肿瘤组织标本进行HE染色比较组织坏死率(HNR)。结果 超声引导下兔肝VX2肿瘤建模成功并在肝内原位成瘤(成功率100%,36/36)。PTX剂量与RNR呈正相关(R^(2)=0.946,P <0.001),HNR呈相似趋势(R2=0.843,P <0.001)。相关分析显示PTX剂量与肿瘤坏死有显著相关,即随着PTX浓度升高,肿瘤的坏死率亦增高(P <0.001)。结论 PTX联合AEI有助于提高治疗兔VX2肝肿瘤的效果。Objective To investigate the success rate of ultrasound-guided modeling of VX2 liver tumors in rabbits and to evaluate the efficacy of paclitaxel(PTX)combined with anhydrous ethanol injection(AEI)in the treatment of rabbit VX2 liver tumors.Methods VX2 tumor cells were injected into the rabbit livers under ultrasound guidance,and tumor growth was observed using contrast-enhanced ultrasound after 2 weeks.The successfully modeled rabbits were divided into 6 groups:the sham surgery group(injected with 2 mL saline),the clinical AEI-only group(AE dosage was determined according to the volume of the tumor),and 4 groups receiving 2 mLAE combined with different concentrations of PTX[PTX in the order of 0 mg(AEI group),12.5 mg(low-dose PTX group),25.0 mg(medium-dose PTX group),and 37.5 mg(high-dose PTX group)].1 week later,contrast-enhanced ultrasound was performed to evaluate tumor necrosis rate(RNR).After euthanizing the animals,tumor tissue specimens were obtained for HE staining to compare tissue necrosis rate(HNR).Results Ultrasound-guided VX2 rabbit liver tumors were successfully modeled,with in situ tumor formation in the liver(success rate 100%,36/36).PTX dosage was positively correlated with the rate of necrosis(R2=0.946,P<0.001),and a similar trend was observed for the high necrosis rate(HNR)(R2=0.843,P<0.001).Correlation analysis showed a significant correlation between PTX dose and tumor necrosis,i.e.,as the PTX concentration increased,the necrosis rate of the tumor also increased(P<0.001).Conclusion PTX combined with AEI can help to improve the efficacy of treating VX2 liver tumors in rabbits.
