摘  要:目的 探讨丝裂原活化蛋白激酶激酶1(MAP2K1)在肾透明细胞癌(cc RCC)组织中的表达及其对ACHN细胞增殖、侵袭和迁移的影响。方法 收集2011年1月至2019年12月国药同煤总医院手术切除的cc RCC组织82例及配对的癌旁组织标本。通过免疫组织化学染色法检测MAP2K1在82例cc RCC癌组织及配对癌旁组织中的表达水平,分析其表达与cc RCC临床病理特征及生存预后的关系。通过CCK-8法、Transwell侵袭和迁移实验检测沉默MAP2K1基因对ACHN细胞生物学功能的影响。结果 癌组织中MAP2K1高表达率高于癌旁组织(74.39%vs23.17%,P<0.001)。癌组织中MAP2K1蛋白相对表达量明显高于癌旁组织[(0.89±0.10) vs(0.21±0.05),P=0.005]。MAP2K1表达水平与TNM分期、T分期相关(P值分别为0.040和0.030)。生存曲线分析显示,MAP2K1高表达患者的中位生存时间为39.3个月,而低表达患者为87.6个月,差异有统计学意义(P<0.001)。沉默MAP2K1基因后,ACHN细胞MAP2K1蛋白表达水平(P=0.037)、72 h细胞增殖活力(P=0.002)、96 h细胞增殖活力(P=0.001)、侵袭力(P=0.014)和迁移力(P=0.002)均降低。结论 MAP2K1在cc RCC癌组织中高表达预示患者不良生存预后,沉默MAP2K1基因可抑制cc RCC癌细胞增殖、侵袭和迁移。MAP2K1可能成为cc RCC的生物学标志物。Objective To investigate the expression of mitogen-activated protein kinase kinase 1(MAP2K1)in clear cell renal cell carcinoma(ccRCC)and its effect on the proliferation,invasion and migration of ACHN cells.Methods From January 2011 to December 2019,ccRCC tissues surgically removed from 82 cases and matched para-cancer tissue specimens at Sinopharm Tongmei General Hospital were collected.Immunohistochemical staining was used to detect the expression level of MAP2K1 in 82 ccRCC cancer tissues and paired adjacent tissues,and to analyze the relationship between MAP2K1 expression and the clinicopathologic features and survival prognosis of ccRCC.The effects of silencing MAP2K1 gene on the biological function of ACHN cells were detected by CCK-8 test,Transwell invasion and migration test.Results The expression rate of MAP2K1 in cancer tissues was higher than that in paracancer tissues(74.39%vs 23.17%,P<0.001).The relative expression of MAP2K1 protein in cancer tissues was significantly higher than that in adjacent tissues[(0.89±0.10)vs(0.21±0.05),P=0.005].The expression level of MAP2K1 was correlated with TNM stage and T stage(P=0.04,P=0.030).The analysis of survival curve showed that the median survival time of patients with high expression of MAP2K1 was 39.3 months,while that of patients with low expression of MAP2K1 was 87.6 months.The difference between the two was statistically significant(P<0.001).After MAP2K1 gene was silenced,MAP2K1 protein expression level(P=0.037),cell proliferation activity at 72 h(P=0.002),cell proliferation activity at 96 h(P=0.001),invasion force(P=0.014)and migration force(P=0.002)of ACHN cells decreased.Conclusions The high expression of MAP2K1 in ccRCC cancer tissue indicates poor survival prognosis of patients.Silencing MAP2K1 gene can inhibit the proliferation,invasion and migration of ccRCC cancer cells.MAP2K1 may become a biological marker of ccRCC.
