摘 要:目的 探讨茶多酚的镇痛抗炎作用关系。方法 购买合格SPF级KM雌性小鼠28只,随机分为4组,即空白对照组(生理盐水)、阳性对照组(阿司匹林肠溶片)、茶多酚低剂量组(50 mg/kg)、茶多酚高剂量组(100 mg/kg),通过热刺痛仪法与醋酸扭体实验诱导的疼痛小鼠模型来开展动物实验,并对血清白介素-1β(IL-1β)、白介素-6(IL-6)及前列腺素E_(2)(PGE_(2))水平进行测定。结果 动物体内实验结果显示,茶多酚能延长热刺痛仪诱导小鼠舔足或抬爪的潜伏时间,提高小鼠的痛阈值(P<0.05),以及减少醋酸诱导的小鼠扭体反应次数(P>0.05);检测指标数据显示,茶多酚能对小鼠血清PGE_(2)的浓度和血清IL-1β水平产生影响(P>0.05),茶多酚不同剂量组IL-6的浓度也减少,其中茶多酚低剂量组显著减少(P<0.05)。结论 茶多酚对疼痛小鼠具有一定的镇痛抗炎药效,且血清PGE_(2)、IL-1β、IL-6水平的降低可能与其镇痛作用机制有关。Objective To explore the analgesic and anti-inflammatory effects of tea polyphenols.Methods Twenty-eight qualified SPF-grade KM female mice were purchased and randomly divided into four groups:blank control group(normal saline),positive control group(aspirin enteric-coated tablets),low-dose tea polyphenol group(50 mg/kg),and high-dose tea polyphenol group(100 mg/kg).Pain models in mice were induced and established using the hot plate test and acetic acid writhing test to conduct animal experiments,and the serum levels of IL-1β,IL-6,and PGE_(2) in the mice were measured.Results The experimental results showed that tea polyphenols could extend the latency of mice′s paw-licking or paw-lifting responses induced by the hot plate test,increasing the pain threshold of the mice(P<0.05),and reduce the number of writhing responses induced by acetic acid(P>0.05).Additionally,tea polyphenols affected the serum concentrations of PGE_(2) and IL-1β in mice(P>0.05).The IL-6 concentration in different dosage groups of tea polyphenols also decreased,with the low-dose tea polyphenol group showing a more significant reduction(P<0.05).Conclusion Tea polyphenols exhibit certain analgesic and anti-inflammatory effects in pain-induced mice,and the reduction in serum levels of PGE_2,IL-1β,and IL-6 in mice may be related to its analgesic mechanism.
