摘　　要：目的探讨锌指蛋白692(ZNF692)作为一种潜在的致癌基因与结肠癌患者预后的相关性。方法应用组织芯片和免疫组织化学(IHC)染色技术,检测ZNF692蛋白在福马林固定、石蜡包埋(FFPE)的248例结肠癌组织中的表达。同时利用R语言对肿瘤基因图谱计划(TCGA)数据库中的176例结直肠癌数据进行分析和验证。利用Wilcoxon秩和检验比较ZNF692在肿瘤组织及正常组织中的表达情况,采用卡方检验分析ZNF692表达与临床病理指标之间的相关性,生存分析采用Kaplan-Meier曲线法及COX多因素回归分析结肠癌患者预后影响因素。结果在肿瘤组织中ZNF692的表达水平远高于邻近正常组织中的表达水平(1.49±0.07比0.27±0.14,P<0.05)。ZNF692低表达组与高表达组在性别、T分期、N分期、神经和/或血管侵犯方面无统计学意义差异。然而,两组在年龄、M分期和分化程度方面差异有统计学意义(P<0.05)。多因素分析显示ZNF692高表达是结肠癌患者不良预后的独立危险因素(P<0.05,HR=1.859,95%置信区间=1.18~3.36),ZNF692低表达组的平均生存月高于高表达组[(47.58±1.36)个月比(38.83±1.57)个月,P<0.05]。结论ZNF692的高表达与结直肠癌患者的年龄、分化程度及远处转移明显相关,ZNF692高表达患者生存期更短。ObjectiveTo investigate the correlation between zinc finger protein 692(ZNF692)as a potential oncogene and the prognosis of patients with colon cancer.MethodsTissue microarray and immunohistochemistry(IHC)staining technique were used to detect the expression of ZNF692 protein in 248 cases of colon cancer formalin-fixed and paraffin-embedded(FFPE)sections.At the same time,the data of 176 cases of colorectal cancer in the cancer genome atlas(TCGA)database were analyzed and verified by R language.Wilcoxon rank sum test was used to compare the expression of ZNF692 in tumor tissues and normal tissues,χ2 test was used to analyze the correlation between the expression of ZNF692 and clinicopathological indexes,and K-M curve method and COX multivariate regression were used to analyze the prognostic factors of colon cancer patients.ResultsThe expression level of ZNF692 in tumor was significantly higher than that in adjacent normal tissues(1.49±0.07 vs.0.27±0.14,P<0.05).There was no significant difference in sex,T stage,N stage,nerve and/or vascular invasion between the low expression group and the high expression group.However,there were significant differences in age,M stage and differentiation between the two groups(P<0.05).Multivariate analysis showed that high expression of ZNF692 was an independent risk factor for poor prognosis of colon cancer patients[P<0.05,HR=1.859,95%confidence interval(CI)=1.18-3.36].The average survival(months)in low ZNF692 expression group was longer than that in high expression group[(47.58±1.36)months vs.(38.83±1.57)moths,P<0.05].Bioinformatics analysis also confirmed that the expression level of ZNF692 affected the prognosis of patients with colorectal cancer.ConclusionThe high expression of ZNF692 is obviously related to the age,differentiation and distant metastasis of colorectal cancer patients,and the survival time of patients with high expression of ZNF692 is shorter.
