重楼皂苷Ⅰ对雄激素非依赖型前列腺癌PC-3细胞增殖与凋亡的影响

摘　　要：目的研究重楼皂苷Ⅰ对雄激素非依赖型前列腺癌PC-3细胞生长抑制与诱导凋亡的作用。方法CCK-8法检测重楼皂苷Ⅰ对PC-3细胞生长的影响;流式细胞仪检测重楼皂苷Ⅰ对PC-3细胞周期的影响;TUNEL法检测重楼皂苷Ⅰ对PC-3细胞凋亡的影响。结果重楼皂苷Ⅰ高、中、低剂量组(1、0.5、0.25μg/mL)能时间和浓度依赖性的抑制PC-3细胞,与阴性对照组比较,抑制率差异有统计学意义(P<0.01);高、中剂量组与阳性对照组(顺铂2.5μg/mL)比较,抑制率差异有统计学意义(P<0.01);流式细胞检测提示:重楼皂苷Ⅰ高、中、低剂量组作用于PC-3细胞后,G0/G1期细胞明显增加,S期细胞减少。与阴性对照组比较,差异有统计学意义(P<0.01)。高、中剂量组与阳性对照组比较,差异有统计学意义(P<0.05);TUNEL法检测提示:重楼皂苷Ⅰ高、中、低剂量组PC-3细胞凋亡率均显著增加,且呈剂量-效果正相关,与阴性对照组比较,差异有统计学意义(P<0.01)。结论重楼皂苷Ⅰ能抑制雄激素非依赖型前列腺癌PC-3细胞的增殖,促进细胞凋亡。Objective To study the effects of PolyphyllinⅠon the growth inhibition and apoptosis induction of androgen independent prostate cancer PC-3 cells.Methods CCK-8 method was used to detect the effect of PolyphyllinⅠon the growth of PC-3 cells.Flow cytometry was used to detect the effect of PolyphyllinⅠon the cell cycle of PC-3 cells.TUNEL method was used to detect the effect of PolyphyllinⅠon apoptosis of PC-3 cells.Results The high,medium,and low dose groups of PolyphyllinⅠ(1,0.5,0.25μG/mL)can inhibit PC-3 cells in a time and concentration dependent manner,with a significant difference in the inhibition rate compared to the negative control group(P<0.01).High and medium dose groups and positive control group(cisplatin 2.5μG/mL),there was a significant difference in the inhibition rate(P<0.01).The flow cytometry test showed that after the high,medium,and low dose groups of Polyphyllin I were applied to PC-3 cells,the G0/G1 phase cells significantly increased,while the S phase cells decreased,with a significant difference compared to the negative control group(P<0.01).There was a significant difference between the high and middle dose groups and the positive control group(P<0.01).The TUNEL assay showed that the apoptosis rate of PC-3 cells in the high,medium,and low dose groups of PolyphyllinⅠwas significantly increased,and there was a dose-effect positive correlation,with a significant difference compared to the negative control group(P<0.01).Conclusion PolyphyllinⅠcan inhibit the proliferation and promote apoptosis of androgen independent prostate cancer PC-3 cells.
