摘 要:目的:研究芍药苷对烫伤大鼠的治疗作用,并基于网络药理学和分子对接技术探讨芍药苷治疗烧伤的作用机制。方法:建立大鼠深Ⅱ度烫伤模型,通过观察芍药苷对大鼠创面外观、计算创面愈合率,创面组织病理学变化;并运用Pubchem、Pahrmmapper、GeneCards等数据库获取芍药苷及烧伤相关靶点,利用STRING数据库和Cytoscape软件,构建并可视化中药(化合物)-靶点-疾病多层次分子网络,通过MCODE分析进行核心基因的筛选,并进行基因本体(GO)及京都基因和基因组百科全书(KEGG)富集分析。最后,利用AutoDock Vina进行分子对接验证。结果:芍药苷能明显促进烫伤大鼠的创面愈合,并促进血管生成及新生上皮形成,减少局部炎症细胞浸润;进一步筛选出芍药苷潜在化学靶点337个,芍药苷与烧伤疾病之间共有靶点138个,主要包括基质金属蛋白酶1(MMP1),肿瘤坏死因子(TNF)等。富集分析结果显示,芍药苷治疗烧伤的核心靶点主要通过PI3K/AKT信号通路、MAPK信号通路等,分子对接计算结果表明,芍药苷与蛋白激酶B1(AKT1)、白蛋白(ALB)、白细胞介素-1β(IL-1β)、TNF、血管内皮生长因子A(VEGFA)等核心靶点有着较好的结合活性。结论:芍药苷通过抑制炎症反应、调控早期微循环、促进血管生成来达到治疗烧伤的目的。本研究证实了芍药苷对烧伤的治疗作用,并揭示相关分子机制。Objective:To investigate the therapeutic effect of paeoniflorin on scald injuries in rats and explore the underlying mechanisms of paeoniflorin in burn treatment based on network pharmacology and molecular docking techniques.Methods:A deep second-degree scald model was established in rats.The effect of paeoniflorin on wound appearance and histopathological changes in wound tissues were observed,and wound healing rate was calculated.Relevant targets of paeoniflorin and burn injuries were identified using databases such as PubChem,PharmMapper,and GeneCards.The STRING database and Cytoscape software were utilized to construct and visualize a multi-level molecular network of Chinese medicine(compound)-target-disease.Core genes were screened through MCODE analysis,followed by Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analyses.Finally,molecular docking validation was performed using AutoDock Vina.Results:Paeoniflorin significantly promoted wound healing in scalded rats,enhanced angiogenesis and new epithelial formation,and reduced local inflammatory cell infiltration.A total of 337 potential chemical targets of paeoniflorin were identified,with 138 common targets related to burn injuries,including matrix metalloproteinase 1(MMP1)and tumor necrosis factor(TNF).Enrichment analysis indicated that the core targets of paeoniflorin in burn treatment were primarily involved in the PI3K/AKT signaling pathway,MAPK signaling pathway,etc.Molecular docking results showed that paeoniflorin exhibited good binding activity with key targets such as protein kinase B1(AKT1),albumin(ALB),interleukin-1β(IL-1β),TNF,and vascular endothelial growth factor A(VEGFA).Conclusion:Paeoniflorin exerts therapeutic effects on burns by inhibiting the inflammatory response,regulating early microcirculation,and promoting angiogenesis.This study confirms the therapeutic potential of paeoniflorin in burn treatment and elucidates the related molecular mechanisms.
