摘 要:目的对水烛香蒲花粉和无苞香蒲的花粉和茎叶中化学成分进行分析,以期探讨香蒲属植物化学成分差异。方法于2022年5—10月,对从南京市建邺区江心洲采集到的两种蒲黄进行了分析。首先通过扫描电镜对两种蒲黄花粉进行显微鉴定,其次采用超高效液相色谱-四极杆飞行时间质谱(UPLC-Q-TOF-MS/MS)进行分析,色谱条件为ACQUITYTM UPLC HSS T3柱(100 mm×2.1 mm,1.8μm),以0.1%甲酸水溶液和乙腈为流动相,梯度洗脱,流速0.3 mL/min,柱温30℃,进样量4μL。通过已知对照品、质谱及文献对化合物进行化学成分分析。结果鉴定出来的两种香蒲分别是水烛香蒲和无苞香蒲。花粉及茎叶中共解析出58个化合物,包含黄酮类27个,有机酸类19个,有机酮类3个,有机酯类3个,聚酮类1个,氨基酸类1个,酰胺类1个,有机醇类1个,苯类1个以及核苷类1个,其中的21个化合物在两种香蒲之间有差异,花粉与茎叶之间也有明显差异,但花粉中黄酮类成分差异不大。结论UPLC-Q-TOF-MS/MS测定方法简便、重现性好,为蒲黄的临床用药与质量标准的建立利用提供了科学依据。Objective To analyze the chemical constituents in the pollen,stem and leaf of Typha angustifolia and Typha laxmannii,with a view to discussing the differences of chemical constituents in the genus Typha.Methods From May to October 2022,two types of Typhae pollen collected from Jiangxinzhou,Jianye District,Nanjing,were analyzed.First,the two types of Typhae pollen were identified by scanning electron microscopy.Second,Ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MS/MS)was used for analysis.The chromatographic condition was ACQUITYTM UPLC HSS T3 column(100 mm×2.1 mm,1.8μm),0.1%formic acid aqueous solution and acetonitrile as mobile phase,gradient elution.The flow rate was 0.3 mL/min,the column temperature was 30℃,and the injection volume was 4μL.Finally,the chemical composition of the identified compounds was analyzed based on reference materials,mass spectrometry,and literature.Results Typha angustifolia and Typha laxmannii were identified.A total of 58 compounds were identified from pollen and stem and leaf,including 27 flavonoids,19 organic acids,3 organic ketones,3 organic esters,1 polyketone,1 amino acid,1 amide,1 organic alcohol,1 benzene and 1 nucleoside,of which 21 compounds were different between the two species,and there were also significant differences between pollen and stem and leaf,but there was no significant difference in flavonoids in pollen.Conclusion The method established in this study is simple and reproducible,which provides a scientific basis for the establishment and utilization of the clinical medication and quality standard of Typha pollen.
