摘  要:为了保护和繁殖地方百合优势变种,以龙牙百合大叶种的鳞片为研究对象,采用植物组织培养技术,研究外植体不同取材部位和HgCl2处理时间的灭菌效果以及不定芽诱导培养、不定芽增殖培养、小鳞茎生根培养的适宜培养基,探讨并建立植株再生体系。结果表明:龙牙百合大叶种进行组织培养时,外植体取材以鳞茎中层鳞片为宜,适宜的灭菌方法为75%乙醇浸泡30 s+30%NaClO处理10 min+0.1%HgCl2浸泡12 min,污染率为12.83%,成活率为85.50%;不定芽诱导适宜培养基为MS+6-BA 1.0 mg/L+NAA 0.3 mg/L时,诱导率为81.17%,分化系数为6.34;不定芽增殖适宜培养基为MS+6-BA 2.0 mg/L+NAA 0.1 mg/L,增殖系数为6.11;小鳞茎生根适宜培养基为1/2 MS+IBA 0.5 mg/L,生根数为11.3条,生根率达到100.00%。In order to protect and propagate the dominant varieties of local lilies,the squamule of Lilium brownii var.viridulum Baker large leaf species were taken as the research object,plant tissue culture technology was used to study the sterilization effects of different parts of explants and different HgCl2 treatment times,as well as the appropriate medium for adventitious bud induction culture,adventitious bud proliferation culture and small bulb rooting culture,and to explore and establish a plant regeneration system.The results showed that in the tissue culture of Lilium brownii var.viridulum Baker large leaf species,the appropriate tissue culture explant was the middle layer squamule of bulbs,and the appropriate sterilization method was 75%ethanol 30 s+30%NaClO 10 min+0.1%HgCl212 min,the contamination rate was 12.83%,and the survival rate was 85.50%.The optimal medium for adventitious bud induction was MS+6-BA 1.0 mg/L+NAA 0.3 mg/L,the induction rate was 81.17%,and the differentiation coefficient was 6.34.The optimal medium for adventitious bud proliferation was MS+6-BA 2.0 mg/L+NAA 0.1 mg/L,the proliferation coefficient was 6.11.The optimal rooting medium for small bulbs was 1/2 MS+IBA 0.5 mg/L,the number of roots was 11.3,and the rooting rate reached 100.00%.
