摘  要:目的通过网络药理学探讨鬼针草治疗肝炎炎症的作用机制。方法用中药系统药理学数据库与分析平台(TCMSP)筛选鬼针草靶点基因;OMIM、Genecards数据库查找肝炎炎症靶点;韦恩在线绘图工具获得共同靶点。利用Cytoscape3.9.1软件以及String数据库构建药物-活性成分-潜在靶点网络图和蛋白质-蛋白质相互作用(PPI)网络图,并进行基因本体(GO)生物过程和京都基因与基因组百科全书(KEGG)通路富集分析。结果筛选出鬼针草6个活性成分和175个潜在靶点,628个肝炎炎症疾病相关靶点,药物与疾病共有60个交集靶点。鬼针草可能通过调节AKT1、TNFRSF1A、IL2等核心靶点,干预IL-17信号通路、TNF信号通路、AGE-RAGE信号通路等相关信号通路,从而达到治疗肝炎炎症的作用。结论鬼针草通过多成分、多靶点、多通路对肝炎炎症具有一定的调控作用。Objective Based on network pharmacology,to explore the mechanism of Bidens bipinnata in treating hepatitis inflammation.Methods The target genes of Bidens japonicum were screened by TCM systematic pharmacology database and analysis platform(TCMSP).OMIM and Genecards databases were used to find hepatitis inflammation targets.Wayne Online drawing tool to obtain common targets.Cytoscape 3.9.1 software and String database were utilized to construct drug-active ingredient-potential target network map and protein-protein interaction(PPI)network map,and gene ontology(GO)biological processes and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis.Results Six active ingredients and 175 potential targets,628 targets related to hepatitis inflammatory diseases,and a total of 60 intersection targets between drugs and diseases were screened.Stipneedling may interfere with IL-17 signaling pathway,TNF signaling pathway,AGE-RAGE signaling pathway and other related signaling pathways by regulating AKT1,TNFRSF1A,IL2 and other core targets,so as to achieve the therapeutic effect of hepatitis inflammation.Conclusion Needleweed has certain regulation effect on hepatitis inflammation through multi-component,multi-target and multi-pathway.
