摘 要:为探明软枣猕猴桃不定根的实际应用,该研究调查了不定根甲醇提取物对枯草芽孢杆菌(Bacillus subtilis)的抑菌活性及其机制。结果表明:提取物对枯草芽孢杆菌抑制效果良好,抑菌圈直径达到21.33 mm,最低抑菌浓度为4 mg·mL^(-1);经提取物处理后的枯草芽孢杆菌的胞外碱性磷酸酶、核酸、蛋白质和电解质水平明显提高,表明提取物破坏了菌细胞膜使其胞内物质渗漏;扫描电子显微镜观察结果显示提取物处理后的枯草芽孢杆菌菌细胞膜皱缩,并有分泌物积聚;提取物抑制枯草芽孢杆菌的呼吸,提取物与磷酸钠的呼吸叠加率(25.0%)较小,推测提取物可能通过调控磷酸戊糖途径(Hexose Monophosphate Pathway,HMP)抑制菌呼吸。进而,对HMP中关键酶之一的葡萄糖-6-磷酸脱氢酶(glucose 6-phosphatedehydrogenase,G6PD)的活性进行了检测,发现提取物处理后G6PD活性降低,从而证明了提取物参与调控HMP。该研究结果可以为今后利用软枣猕猴桃不定根生产抑菌相关产品提供参考。In order to explore the practical application of Actinidia arguta Sieb.et Zucc.adventitious roots,the study investigated the bacteriostatic activity of methanol extract from Actinidia arguta Sieb.et Zucc.adventitious roots against Bacillus subtilis and its mechanism.The results showed that the extracts had good inhibitory effect on Bacillus subtilis,with a 21.33 mm inhibitory circle diameter and a minimum inhibitory concentration of 4 mg·m^(-1).The extracellular alkaline phosphatase,nucleic acids,proteins and electrolyte levels of Bacillus subtilis treated by the extract were significantly increased,indicating that the extract destroyed the cell membrane and caused the leakage of intracellular substances.Scanning electron microscopy results showed that the cell membrane of Bacillus subtilis treated by the extract was wrinkled and secretions were accumulated.The extract inhibited the respiration of Bacillus subtilis,and the respiratory superposition rate of the extract and sodium phosphate(25.0%)was small,supposing that the extract might inhibit bacterial respiration through the regulation of hexose monophosphate pathway(HMP).Furthermore,the activity of glucose-6-phosphate dehydrogenase(G6PD),one of the key enzymes in HMP,was detected,and it was found that the activity of G6PD was decreased after the treatment of the extract,which proved that the extract was involved in the regulation of HMP.The results of this study can provide reference for the use of Actinidia arguta Sieb.et Zucc.adventitious roots to produce the bacteriostatic products in the future.
