摘 要:为探究非编码RNA对鹅骨骼肌生长发育的调控机制,试验以20胚龄和孵化后3日龄马岗鹅的腿部肌肉作为研究对象,通过HE染色观察不同阶段腿肌组织结构差异,并通过RNA测序探讨可能影响腿肌生长发育的RNAs。3日龄马岗鹅腿肌相较于20胚龄显示出了完整的肌纤维结构,肌纤维轮廓清晰;两阶段内共有2289个差异表达mRNAs,17个差异表达miRNAs和192个差异表达lncRNAs;差异表达基因在骨骼肌生长和发育相关的通路中显著富集,如细胞周期、FoxO信号通路和MAPK通路等;研究通过预测差异表达的mRNA和lncRNA与miRNA的靶向关系,成功构建一个与骨骼肌生长发育密切相关的ceRNA网络,其中36个lncRNAs、10个miRNAs被预测可以调控CCNB2、SHC4、ANGPT2、NRK、SLC25A4、WHAMM、FREM1、HMMR、PHKG1、DUSP1和COL1A2等与腿肌生长发育相关的基因。研究结果揭示了lncRNA和miRNA在鹅腿肌生长和发育中的潜在功能和机制,为进一步的研究提供了参考。To investigate the regulatory mechanisms of non-coding RNAs in goose skeletal muscle growth and development,this study focused on the leg muscles of 20-day-age embryos and 3-day-age post-hatched Magang goose.Histological examina⁃tion using HE staining was performed to observe the differences in muscle fiber between different stages,and RNA sequencing was conducted to explore differentially expressed RNAs that might affect leg muscle growth and development.The results showed that the leg muscle of 3-day-age Magang goose exhibited a well-formed muscle fiber structure with clear contours compared to the 20-day-age embryos;There were 2289 differentially expressed mRNAs,17 differentially expressed miRNAs and 192 differ⁃entially expressed lncRNAs in the two stages,and the significant enrichment of differentially expressed genes were in pathways re⁃lated to skeletal muscle growth and development,including cell cycle,FoxO signaling pathway and MAPK pathway.The study successfully constructed a ceRNA network closely associated with skeletal muscle growth and development by predicting the tar⁃geted relationship between differentially expressed mRNA,lncRNA and miRNA.Among them,36 lncRNAs and 10 miRNAs were predicted to regulate genes associated with leg muscle growth and development,including CCNB2,SHC4,ANGPT2,NRK,SLC25A4,WHAMM,FREM1,HMMR,PHKG1,DUSP1 and COL1A2.These findings revealed the potential functions and mecha⁃nisms of lncRNAs and miRNAs in goose leg muscle growth and development,provided references for further research.
