摘 要:给予15月龄BALB/c小鼠高牛肉蛋白饲料,并以消食片和不同剂量的桑叶粉和魔芋粉组合物(以下简称组合物)干预,评价组合物对高牛肉蛋白饮食老年小鼠蛋白质代谢的作用。结果表明,组合物干预提高动物摄食量、粪便和盲肠内容物含水量,蛋白质表观消化率高于92.92%,较高牛肉蛋白饮食组提高6.22%以上(P<0.05),粪便pH值降低。血清中谷草转氨酶、谷丙转氨酶活性和氨基酸水平均提高,血清尿素氮、粪便氨基酸含量降低。粪便多肽结果分析表明,肽段分子质量主要分布在1000~1500 Da,组合物干预使各组特有肽段数减少。小肠质构和组织测量发现,组合物干预使小肠坚实度、弹性、断裂力和断裂距离分别提高了14.49%、10.38%、41.92%和7.88%,绒毛高度/隐窝深度值显著增大(P<0.05),说明组合物改善了老年小鼠小肠结构,与小肠苏木精-伊红染色观察结果一致。组合物通过改善肠道结构促进了高蛋白饮食老年小鼠蛋白质消化代谢。This intervention aimed to assess the mulberry leaf powder and konjac powder composition(hereinafter referred to as'composition')impact on protein metabolism in aged mice consuming a high beef protein diet.Fifteen-month-old BALB/c mice were administered a high beef protein diet and subjected to intervention with the combination or digestible tablets.The study revealed that the intervention led to increased food intake,higher water content in fecal and cecal contents,enhanced protein apparent digestibility to 92.92%,higher 6.22%than HBP group(P<0.05),and a decrease in fecal pH value.Moreover,the intervention resulted in elevated activity and levels of glutamic-oxalacetic transaminase(GOT)and glutamic-pyruvic transaminase(GPT)in serum,alongside reduced levels of serum urea nitrogen and fecal amino acids.Analysis of fecal peptides indicated a predominant molecular weight distribution of peptide segments between 1000 Da and 1500 Da,with the intervention reducing the number of unique peptide segments in each group.The texture and tissue measurements of the small intestine revealed notable enhancements following the combined intervention,with increases of 14.49%in firmness,10.38%in elasticity,41.92%in breaking force,and 7.88%in breaking distance.Moreover,the ratio of villus height/crypt depth exhibited a significant increase(P<0.05),indicative of structural improvements in the small intestine due to the intervention.This finding aligns with the findings from the small intestine hematoxylin-eosin observations.The combination intervention enhances protein digestion and metabolism in elderly mice on a high protein diet by improving intestinal structure.
