摘  要:研究大孔树脂对黑果腺肋花楸果中多种活性成分同时纯化工艺条件及各组分进行体外活性,并考察在体外模拟胃肠条件下总皂苷的释放量。采用静态、动态的吸附与解吸方法进行纯化工艺研究,采用UVVis进行各组分的体外活性研究。最佳纯化工艺条件为:LSA-10大孔树脂,生药浓度48.0 mg/mL,吸附流速1.0 mL/min,洗脱流速1.5 mL/min。通过水、20%和80%乙醇水溶液的洗脱得到多组分。纯化后干浸膏中多糖、花色苷、总黄酮和总皂苷的含量分别为615.20、127.55、48.97、797.49 mg/g。体外活性实验表明各组分均具有不同活性的IC50值。该工艺条件下的大孔树脂可以同时有效纯化多种活性成分,所得各组分均具有不同的体外活性,胃肠消化酶可促进总皂苷释放量。本研究为黑果腺肋花楸果的开发利用提供了数据参考。This study investigated the simultaneous purification and in vitro activities of multiple active components in the black chokeberry fruits using macroporous resin.The release of total saponins under simulated gastrointestinal conditions was also evaluated.Static and dynamic adsorption and desorption methods were employed for purification process,while UV-Vis was used for in vitro activity studies of each component.The optimal purification conditions were as follows:LSA-10 macroporous resin,crude drug concentration of 48.0 mg/mL,adsorption flow rate of 1.0 mL/min,and elution flow rate of 1.5 mL/min.Multiple components were obtained through elution with water,20%,and 80%ethanol aqueous solutions.After purification,the contents of polysaccharides,anthocyanins,total flavonoids,and total saponins in the dry extract were 615.20,127.55,48.97,and 797.49 mg/g,respectively.In vitro activity experiments showed that each component had different IC50 values.Under these conditions,the macroporous resin could effectively purify multiple active components simultaneously,and the obtained components exhibited distinct in vitro activities.Gastrointestinal digestive enzymes were found to promote the release of total saponins.The study provides data reference for the development and utilization of black chokeberry fruits.
