摘  要:使用高效液相色谱法(HPLC)测定肉桂挥发油中肉桂醛、肉桂醇、苯甲醛、肉桂酸等4种化学成分含量,采用白色封闭群(Wistar)大鼠灌胃给与肉桂醛,测定肉桂醛在大鼠体内的基本代谢参数。蒸馏法提取肉桂挥发油的得率为3.03 mL/g,肉桂挥发油中肉桂醛、苯甲醛、肉桂酸、肉桂醇平均含量分别为50.13%、0.64%、0.45%、3.29%。试验大鼠单次灌胃100、200、400 mg/kg剂量肉桂醛后,吸收速率常数Ka分别为20.187、18.708、20.437 min,表明肉桂醛能被迅速吸收;灌胃给药后血浆中肉桂醛在50 min即达峰浓度值(C_(max))=19.980、38.806、90.106 mg/L,达峰时间(T_(max))均在0.833h左右,半衰期(T_(1/2))为1.7h。表明肉桂醛进入大鼠体循环后,分布迅速,并快速代谢为肉桂酸,无明显的蓄积作用,其吸收和消除与给药剂量无关,代谢过程符合开放的二室模型。High performance liquid chromatography(HPLC)was used to determine the contents of cinnamaldehyde,cinnamyl alcohol,benzaldehyde and cinnamic acid in the volatile oil of cinnamomum cassia.Cinnamaldehyde was administrated by gavage to Wistar rats,and basic metabolic parameters of cinnamaldehyde in rats was determined.The yield of cinnamon volatile oil extracted by distillation was 3.03 mL/g.Besides,the average contents of cinnamaldehyde,benzaldehyde,cinnamic acid,and cinnamyl alcohol in cinnamon volatile oil were 50.13%,0.64%,0.45%,and 3.29%,respectively.Rats given a single oral dose of 100,200,or 400 mg/kg cinnamaldehyde showed absorption rate constants(Ka)of 20.187,18.708,and 20.437 min respectively,indicating rapid absorption of cinnamaldehyde in the rats gastrointestinal tract.Peak plasma concentrations of cinnamaldehyde(C_(max))after oral administration at doses of 100,200,and 400 mg/kg were 19.980,38.806,and 90.106 mg/L at 50 minutes respectively,with peak times(T_(max))around 0.833 hours and a half-life(T_(1/2))of 1.7 hours.This suggested rapid distribution of cinnamaldehyde in rat circulation,quick metabolism to cinnamic acid without significant accumulation,and its absorption and elimination being independent of the dose,following an open two-compartment model.
