结直肠侧向发育型肿瘤可能发病机制研究

摘  要:目的:结直肠侧向发育型肿瘤(LST)检出率逐年升高,但其发病机制尚不明确。本研究将通过生物信息学方法分析该疾病mRNA表达数据集,探讨其发病机制。方法:利用差异基因分析,明确GSE77635中LST与正常对照组差异基因表达的情况,并对差异基因进行GO及KEGG分析。利用差异基因建立蛋白相互作用网络后选取关键基因进行分析。结果:共有1194个差异基因,其中包括469个上调基因及725个下调基因。其中PI3K-Akt信号通路可能参与LST的发生及发展。建立PPI网络后共选取5个关键差异基因(TP53、FN1、MYC、CD44、KRAS)。结论:LST的发生可能与PI3K-Akt通路密切相关。Objective:The detection rate of lateralized developmental colorectal tumors(LST)is increasing year by year,but its pathogenesis is unclear.In this study,we will analyze the mRNA expression dataset of this disease by bioinformatics methods to explore its pathogenesis.Methods:Differential gene analysis was used to clarify the differential gene expression of LST in GSE77635 versus normal controls,GO and KEGG analyses were performed on the differential genes.Key genes were selected for analysis after establishing protein interaction networks using differential genes.Results:There were 1194 differential genes,including 469 up-regulated genes and 725 down-regulated genes.Among them,PI3K-Akt signalling pathway may be involved in the development of LST.A total of 5 key differential genes(TP53,FN1,MYC,CD44,KRAS)were selected after establishing the PPI network.Conclusion:The occurrence of LST may be closely related to the PI3K-Akt pathway.
