摘  要:以体外清除羟自由基作为活性筛选模型,首先将木棉花瓣95%乙醇提取物分为不同极性部分,比较其清除羟自由基活性,然后将活性较好的部位采用柱层析法初步分离,比较各洗脱物对羟自由基的清除作用,最后将活性较好的洗脱物采用制备薄层层析法纯化得到单体,测试其在不同浓度下对羟自由基的清除作用并鉴定结构类型。实验结果表明木棉花瓣醇提取物中对羟自由基清除作用较强的是无水乙醇溶解部分,该部分经柱层析初步分离共得到12个洗脱物,其中6个洗脱物的清除率均大于80%,采用制备薄层层析法从中分离纯化得到七个单体,其中三个单体的量较多,对羟自由基的清除作用与浓度呈正相关性,二个清除率较大的单体经鉴定结构类型为黄酮类。Using in vitro hydroxyl radical scavenging as an activity screening model,different polar parts were obtained from the 95%ethanol extract of the petal of Bombax ceiba L.and their activities in scavenging hydroxyl radical were compared.Then the part with better activity effect was separated into different eluates preliminarily by the method of column chromatography(CC)and the activities of these eluates on scavenging hydroxyl radical were compared.At last,the eluates which had better activities than the others were purified to obtain monomers by preparative thin layer chromatography(PTLC).The concentration correlation of these monomers on scavenging hydroxyl radical was studied and the type of the structures of the active compounds were identified.The results showed that the anhydrous ethanol dissolved part which was obtained from the 95%ethanol extract of the petal of Bombax ceiba L.had better activities than the others.Then 12 eluates were obtained from this part by CC and their activities on scavenging hydroxyl radical were compared,in which the clearance rates of 6 eluates were greater than 80%.Seven main monomers were obtained from these eluates by the methods of PTLC,in which three monomers had strong activities on scavenging hydroxyl radical and their scavenging rates were related to the concentrations.Two main monomers were identified as flavonoids.
