摘 要:【目的】旨在研究2种谷氨酰胺二肽对奶牛小肠上皮细胞增殖及凋亡的影响。【方法】对体外培养的奶牛小肠上皮细胞进行谷氨酰胺(Glutamine,Gln)饥饿处理后,分别添加Gln、丙氨酰谷氨酰胺(Alanyl-glutamine,Ala-Gln)和甘氨酰谷氨酰胺(Glycyl-glutamine,Gly-Gln)进行处理,检测各处理组细胞的细胞活力、细胞凋亡率及周期、乳酸脱氢酶(lactic dehydrogenase,LDH)漏出量及凋亡相关蛋白(Bax、Bcl-2和Caspase-3)的表达量。【结果】与CK(Gln饥饿处理组)相比,添加Ala-Gln或Gly-Gln的奶牛小肠上皮细胞的细胞活力显著提高(P<0.05),LDH漏出量显著降低(P<0.05)。此外,与CK相比,添加Gln、Ala-Gln及Gly-Gln均能显著降低G0/G1和G2/M期阻滞细胞比率,提高S期细胞比率(P<0.05),也能显著提高抗凋亡因子Bcl-2蛋白表达,而显著降低促凋亡因子Bax和Caspase-3蛋白表达(P<0.05),从而显著降低细胞凋亡率(P<0.05)。【结论】谷氨酰胺二肽(Ala-Gln及Gly-Gln)能够促进奶牛小肠上皮细胞增殖并减缓其凋亡,从而保护肠道黏膜结构的完整性。该研究结果可为谷氨酰胺二肽在奶牛肠道营养中的应用提供理论依据。【Objective】This study is conducted to investigate the effects of glutamine dipeptide on the proliferation and apoptosis of bovine small intestinal epithelial cells.【Method】Bovine small intestinal epithelial cells were cultured in vitro and subjected to Glutamine(Gln)starvation treatment.Gln,Alanyl-glutamine(Ala-Gln),or Glycine-glutamine(Gly-Gln)were added respectively.Cell viability,rate and cycle of apoptosis,overflow amount of lactic dehydrogenase(LDH),and expression levels of apoptotic-related proteins(Bax,Bcl-2,and Caspase-3)were measured.【Result】The results showed as follows:compared with CK(the Gln starvation group),the bovine small intestinal epithelial cells supplemented with Ala-Gln and Gly-Gln significantly increased cell viability and significantly decreased LDH amount(P<0.05).In addition,compared with CK,the addition of Gln,Ala-Gln and Gly-Gln significantly decreased the rate of G0/G1 and G2/M phase retarded cells and increased the rate of S phase cells(P<0.05);The apoptosis rate was significantly decreased(P<0.05);The expression of anti-apoptotic factor Bcl-2 was significantly increased,while the expressions of pro-apoptotic factor Bax and Caspase-3 were significantly decreased(P<0.05).【Conclusion】In conclusion,glutamine dipeptides(Ala-Gln and Gly-Gln)can promote the proliferation and reduce the apoptosis of bovine small intestinal epithelial cells,thus protecting the integrity of the intestinal mucosal structure,which can provide a theoretical basis for the application of glutamine dipeptide in bovine intestinal nutrition.
