橄榄苦苷对人骨肉瘤MG-63细胞的增殖抑制和诱导凋亡

摘　　要：目的研究橄榄苦苷对人骨肉瘤MG-63细胞的活性的影响。方法PBS溶解配置不同浓度的橄榄苦苷(0、25、50、100、200、400μg/mL)处理MG-63细胞24 h或48 h,CCK8、克隆形成实验分析细胞的活性及增殖能力的变化;Annexin-V/PI双染法分析细胞的凋亡水平。结果橄榄苦苷处理后,MG-63细胞活性及增殖能力显著降低,且凋亡水平显著增加(P<0.05),48 h的IC 50低于24 h的IC 50,对肿瘤细胞的抑制作用与橄榄苦苷的浓度和处理时间成正比(P<0.05)。结论橄榄苦苷对人骨肉瘤细胞株MG-63的活性和增殖能力具有抑制能力,能够诱导MG-63的凋亡。Objective To study the effect of oleuropein on the activity of human osteosarcoma MG-63 cells.Methods MG-63 cells were dissolved in Polybutylene Succinate(PBS)and prepared with different concentrations of oleuropein(0,25,50,100,200,400μg/ml)to treat MG-63 cells for 24h or 48h,and the changes in cell activity and proliferation ability were analyzed by cell counting Kit-8(CCK8)and clone formation assays.Annexin-V/PI double staining method was performed to analyze the cells apoptosis level.Results After treatment with oleuropein,the activity and proliferation of MG-63 cells were significantly reduced,and apoptosis level was significantly increased(P<0.05).The 48-h inhibitory concentration 50%(IC50)was significantly lower than 24-h IC50.The inhibitory effect on tumor cells was positively correlated with the concentration and treatment time of oleuropein(P<0.05).Conclusion Oleuropein can significantly inhibit the activity and proliferation of human osteosarcoma cell line MG-63,which can induce MG-63 apoptosis.
