摘 要:利用纯培养技术从活性污泥中分离筛选出具有高效产电功能的细菌HAUT-1,利用16S rRNA基因序列比对分析对其加以鉴定并应用于MFCs中进行产电能力测试及单因素条件优化。结果表明,菌株HAUT-1的16S rRNA序列与Corynebacterium vitarumen(X84680)的同源性最高为98%,因此将该菌株命名为Corynebacterium vitarumen HAUT-1。单因素实验分析结果表明,该菌株在MFCs中的最佳产电条件是以乙酸钠作为碳源,甘氨酸作为氮源,培养pH为7,培养温度为25℃,在该条件下,菌株HAUT-1在MFCs中的最大输出电压可以达到635 mV;进一步的极化和功率密度曲线分析结果表明,MFCs系统的开路电压为608 mV,当电流密度为24 mA/cm^(2)时,输出功率密度达到最大,为53.88 mW/cm^(2)。结果表明,筛选出的菌株HAUT-1具有稳定且高效的产电能力,对产电微生物菌种资源开发,进一步提升MFCs效率,同步实现环境修复和产能输出具有重要意义。The basic isolated and screened a novel strain HAUT-1 from activated sludge using pure culture technology,identified it by 16 S rRNA gene sequence analysis,and applied it in MFCs for power generation testing and single-factor condition optimization.The results showed that the 16S rRNA sequence of strain HAUT-1 had the highest homology of 98%with Corynebacterium vitarumen(X84680),and thus the strain was named Corynebacterium vitarumen HAUT-1.The results of the single-factor test showed that the optimal power output conditions for this strain in MFCs were sodium acetate as the carbon source,glycine as the nitrogen source,incubation pH of 7,and incubation temperature of 25℃,under which the maximum output voltage of strain HAUT-1 in MFCs could reach 635 mV;further analysis of polarization curves and power density curves showed that the open-circuit voltage of the MFCs system was 608 mV,and the output power density reached the maximum of 53.88 mW/cm^(2)when the current density was 24 mA/cm^(2).The results showed that the screened strain HAUT-1 in this study has a stable and highly efficient power output capacity,which is of great significance for the development of electrogenic strain resources to further enhance the efficiency of MFCs and realize the dual effects of environmental remediation and power output.
