摘 要:目的探究梓醇对甲基苯骈三氮唑(MPTP)诱导帕金森病模型小鼠的神经保护作用,并探索其作用机制。方法利用MPTP构建帕金森病模型小鼠模型,根据实验目的将实验小鼠分成空白对照组,MPTP模型组和MPTP+梓醇组。利用开放场实验(OFT)、爬杆实验和Rota-rod实验检测小鼠行为学,处死小鼠后收集其脑组织,检测小鼠脑组织中活性氧(ROS)含量和超氧化物歧化酶(SOD)活性,Western blot法检测凋亡相关分子的表达量,免疫组织化学检测小鼠脑黑质和纹状体中酪氨酸羟化酶(HT)的表达。结果3组小鼠的OFT总距离,爬杆试验结果和Rota-rod时间差异均无统计学意义(均P>0.05),但MPTP+梓醇组小鼠的区域中心距离和进入竞技场中心的次数均优于MPTP组(均P<0.05)。MPTP组小鼠脑组织中ROS含量高于空白对照组(P<0.05),而SOD活性低于空白对照组(P<0.05);MPTP+梓醇组小鼠脑组织中ROS含量低于MPTP组(P<0.05),而SOD活性高于MPTP组(P<0.05)。MPTP模型组小鼠Cleaved-Caspase-9和Cleaved-Caspase-3的相对表达量均高于空白对照组(均P<0.05);MPTP+梓醇组小鼠的Cleaved-Caspase-9和Cleaved-Caspase-3的相对表达量均低于MPTP组(均P<0.05)。MPTP+梓醇组小鼠的p-MKK4/MKK4、p-JNK/JNK和p-cjun/jun的相对表达量均低于MPTP组(均P<0.05)。MPTP模型组小鼠黑质和纹状体中HT的含量均低于空白对照组(均P<0.05),而MPTP+梓醇组小鼠黑质和纹状体中HT的含量均高于MPTP模型组(均P<0.05)。结论梓醇不仅可以抑制氧化反应,还可以调高HT的表达,进而发挥抗帕金森的作用,而MKK4/JNK/c-jun信号通路可能是MPTP氧化应激传导的信号通路。Objective To explore the neuroprotective effect ofCatalpol on MPTP induced Parkinson's disease mod-el mice,and analyse its mechanism.Methods Parkinson's disease model mice were made by MPTP,and dividedinto blank group,MPTP model group and MPTP+Catalpol group.The open field test(OFT),Pole climbing test,andRota-rod test were used to observe the behavior ofthe mice.Killed the mice,collected their brain tissue,and detectedthe content ofreactiveoxygen species(ROS).The activity ofsuperoxide dismutase(SOD),theexpression ofapoptosis related molecules was detected by Western blot.The expression oftyrosine hydroxylase(HT)in substantia nigraand striatum were detected by immunohistochemistry.ResultsThere were no significant differences in the total dis-tance traveled.poleclimbing times.and Rota-rod times among the three groups(all P>0.05).MPTP+Catalpol grouphad abtter distance to the center area and ahigher numberofentries into the centerofthe arena compared to MPTPgroup(P<0.05).The content of ROS in MPTP group was significantly higher than that in the blank group(P<0.05),the activity ofSOD was significantly lower than that in MPTP group(P<0.05).The content ofROS in MPTP+Catalpol group was significantly lower than that in MPTP group(P<0.05),the activity of SOD was significantlyhigher than that in MPTP group(P<0.05).The relative expression of cleaved-caspase-9 and cleaved-caspase-3 in MPTP model group were significantly higher than those in the blank group(all P<0.05),while MPTP+Catalpol groupwere significantly lower than those in MPTP group(all P<0.05).The relative expression levels of p-MKK4/MKK4.p-JNK/JNK,and p-cjun/jun in MPTP+Catalpol group were significantly lower than those in MPTP group(all P<0.05)The content ofHT in substantia nigra and striatum ofMPTPmodel group were significantly lower than those in the blankgroup(all P<0.05),while MPTP+Catalpol group were significantly higher than those in MPTP model group(all P<0.05).Conclusions Catalpol can not only inhibit the oxidative reaction,but also increase the expression
