摘 要:目的探讨川芎活性物质阿魏酸乙酯(EFE)联合有氧运动(AE)对阿尔茨海默病(AD)大鼠认知功能的影响及作用机制。方法将70只SD大鼠随机分为5组(n=12):对照组、模型组、AE组、EFE组和AE+EFE组。通过对大鼠双侧海马注射β-淀粉样蛋白1-42(Aβ1-42)诱导AD大鼠模型。建模1周后,AE组和AE+EFE组大鼠进行有氧跑台运动,其他组大鼠不运动;EFE组和AE+EFE组大鼠灌胃2 mL 100 mg/kg的阿魏酸乙酯,其他组大鼠灌胃2 mL 0.5%羧甲基纤维素钠,共灌胃6周。给药结束后,通过Morris水迷宫实验评估大鼠的认知功能。根据试剂盒说明检测海马组织超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和丙二醛(MDA)水平。利用qRT-PCR检测海马组织β-淀粉样前体蛋白裂解酶1(BACE1)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、核因子红细胞2相关因子2(Nrf2)、Kelch样环氧氯丙烷相关蛋白-1(Keap1)和血红素加氧酶1(HO-1)转录水平。通过Western blot检测海马组织BACE1和Aβ1-42蛋白表达水平。结果与AE组和EFE组比较,AE+EFE组的逃避潜伏期降低,60 s内穿越平台的次数显著升高(P<0.05),海马组织SOD和CAT水平显著升高,海马组织MDA水平显著降低(P<0.05)。与AE组和EFE组比较,AE+EFE组的海马组织TNF-α、IL-1β、IL-6和Keap1mRNA相对表达量显著降低(P<0.05),而Nrf2和HO-1 mRNA相对表达量显著升高。与AE组和EFE组比较,AE+EFE组的海马组织BACE1 mRNA、BACE1蛋白和Aβ1-42蛋白相对表达量降低(P<0.05)。结论阿魏酸乙酯联合有氧运动可有效改善AD大鼠的认知功能,其机制可能与抑制氧化应激、炎症和Aβ的积累有关。Objective To investigate the effect of combination of Ligusticum chuanxiong active substance ethyl ferulate(EFE)and aerobic exercise(AE)on cognitive function of rats with Alzheimer’s disease(AD).Methods Seventy SD rats were randomly divided into 5 groups(Control group,Model group,AE group,EFE group and AE+EFE group n=12).The rat model of AD was induced by injectingβ-amyloid 1-42(Aβ1-42)into the hippocampus of rats on both sides.One week after modeling,the rats in AE group and AE+EFE group took aerobic treadmill exercise,while the other groups did not exercise.And the rats in EFE group and AE+EFE group were gavaged with 2 mL 100 mg/kg ethyl ferulate,the rats in other groups were gavaged with 2 mL 0.5%sodium carboxymethyl cellulose.All groups were treated for 6 weeks.After administration,the rats’cognitive function was assessed by the Morris Water maze test.The levels of superoxide dismutase(SOD),catalase(CAT)and malondialdehyde(MDA)in hippocampal tissue were detected according to the instructions of the kit.Hippocampalβ-amyloid precursor protein lyase 1(BACE1),tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6),nuclear factor erythrocyte 2-associated factor 2(Nrf2),Kelch-like ECHassociated protein 1(Keap1)and heme oxygenase 1(HO-1)transcription levels were detected by qRT-PCR.The expression levels of BACE1 and Aβ1-42 in hippocampus were detected by western blot.Results Compared with AE group and EFE group,the escape latency of AE+EFE group decreased,and the number of crossing the platform within 60 s increased significantly(P<0.05).Compared with AE group and EFE group,SOD and CAT levels in the hippocampus of AE+EFE group increased,while MDA levels in the hippocampus decreased significantly(P<0.05).Compared with AE and EFE groups,the mRNA relative expressions of TNF-α,IL-1βand IL-6 in hippocampus of AE+EFE group decreased significantly(P<0.05).Compared with AE and EFE groups,the relative expression levels of Nrf2 and HO-1 mRNA in the hippocampus of AE+EFE group were increased,whil
