摘 要:目的:探讨活化的巨噬细胞对大鼠晶状体损伤后视神经损伤修复的影响。方法:选用普通级成年健康Wistar大鼠45只,随机平均分为3组:正常对照组不做任何处理;NC组给予视神经横断性损伤;NC+LP组先给予视神经横断性损伤,再给予晶状体损伤。术后第7、14、21天每组分别处死大鼠5只,免疫组化染色方法检测视网膜组织中ED-1的表达,免疫荧光法检测GAP-43的表达。结果:术后第7、14、21天,正常对照组及NC组视网膜中均未见ED-1阳性细胞;NC+LP组ED-1阳性细胞计数分别为(58.27±4.79)、(45.39±5.13)、(21.17±3.40),第7天时表达最强,之后降低(P<0.001)。术后第7天NC组神经节细胞层可见少量GAP-43表达,高于正常对照组,之后表达降低;NC+LP组各时间点GAP-43表达均大于NC组(P<0.05),术后第14天表达最强(P<0.05)。结论:激活巨噬细胞可促进晶状体损伤后视神经轴突再生。Aim:To investigate the effects of activated macrophages on the regeneration of optic nerve axotomy after lens injury of rat.Methods:A total of 45 healthy adult Wistar rats were randomly allocated into 3 groups,15 in each group:normal control group(without any treatment),NC group(with optic nerve axotomy),NC+LP group(with optic nerve axotomy firstly,and then lens injury).On the 7th,14th,21th day after operation,5 rats were executed,respectively,in each group,ED-1 expression and GAP-43 expression in retinal tissue were detected by immunohistochemistry and immunofluorescence staining,respectively.Results:On the 7th,14th,21th day after operation,ED-1 positive cells were not detected in normal control group and NC group;while the number of ED-1 positively cells was(58.27±4.79),(45.39±5.13),(21.17±3.40)in NC+LP group,which was the highest at the 7th day,and gradually decreased over time(P<0.001).On the 7th day,the expression of GAP-43 in retinal ganglion cell layer of NC group was higher than that of normal control group(P<0.05),and then decreased;the expression of GAP-43 RGC of NC+LP group was higher than that of NC group at all time points(P<0.05),and the expression was strongest on the 14th day after operation(P<0.05).Conclusion:The activation of macrophages could promote axonal regeneration of the optic nerve after lens injury.
