摘 要:目的探讨人参皂甙Rg3抑制卵巢癌细胞生长可能存在的分子机制。方法采用细胞克隆实验检测人参皂甙Rg3对人卵巢癌SKOV3细胞增殖的抑制效果;四氮唑蓝(MTT)比色法、流式凋亡、划痕实验、Transwell实验分别检测人参皂甙Rg3对SKOV3细胞凋亡、侵袭、迁移的影响;qRT-PCR法和Western blot法分别检测Rg3不同浓度作用下卵巢癌细胞中Zeste基因增强子同源物2(EZH2)基因和蛋白的表达。结果人参皂甙Rg3对SKOV3细胞增殖抑制率升高(P<0.05),凋亡率显著上升(P<0.01),侵袭、迁移能力均显著下降(P<0.01),SKOV3细胞中EZH2的基因和蛋白随着Rg3浓度的增加表达量呈现明显的下降(P<0.01)。结论人参皂甙Rg3可能通过抑制EZH2的表达,促进卵巢癌细胞的凋亡,抑制卵巢癌SKOV3细胞增殖、侵袭以及迁移的能力,为卵巢癌的临床治疗提供新思路。Objective To investigate the possible molecular mechanism by which ginsenoside Rg3 inhibits the growth of ovarian cancer cells.Methods Human ovarian cancer cells SKOV3 were studied.The inhibitory effect of ginsenoside Rg3 on SKOV3 cell proliferation was detected by cell cloning assay.The effects of ginsenoside Rg3 on apoptosis,invasion and migration of SKOV3 cells were detected by MTT colorimetry,flowcytometry,scratch assay and Transwellassay,respectively.qRT-PCR and Western blot were used to detect the expression of enhancer of zeste homolog 2(EZH2)gene and protein in ovarian cancer cells underdifferent concentrations of Rg3.Results Ginsenoside Rg3 increased the proliferation inhibition rate of SKOV3 cells(P<0.05),increased the apoptosis rate(P<0.01),and decreased the invasion and migration abilities(P<0.01).The expression of EZH2 gene and protein in SKOV3 cells decreased significantly with the increase of Rg3 concentration(P<0.01).Conclusion Ginsenoside Rg3 may promote the apoptosis of ovarian cancer SKOV3 cells and inhibit the proliferation,invasion and migration of the ovarian cancer cells by inhibiting the expression of EZH2,providing new ideas for the clinical treatment of ovarian cancer.
