摘  要:【目的】探明不同地区薜荔叶片类黄酮种类和含量的差异,为薜荔的品质评价标准制订、种质资源保护和开发利用提供参考。【方法】从海南文昌、海南定安、广东广州、福建漳州采集薜荔叶片样本,对叶片进行类黄酮成分粗提后,利用定量代谢组学技术对其定性定量分析;在此基础上,通过PCA、热图聚类分析、基于OPLS-DA的差异代谢物筛选和KEGG富集分析,研究不同地区薜荔叶片类黄酮的代谢差异。【结果】鉴定获得14类96种类黄酮化合物,其中,黄酮和黄酮醇的种类较多,各有24种;来自定安的薜荔类黄酮种类和含量最丰富,分别为77种和2.15μmol/g。4个地区样本的(-)-儿茶素丰度均最高,分别为1.45μmol/g(定安)、1.23μmol/g(漳州)、1.20μmol/g(广州)、1.04μmol/g(文昌);芦丁、(-)-表儿茶素和山奈酚-3-O-芸香糖苷在总黄酮中占比较大,部分化合物仅在特定地区样本中富集,如木犀草苷仅在文昌薜荔样本中被检测到。不同地区样本类黄酮的积累模式差异较大,具有区域特异性。差异代谢物主要集中在代谢途径、黄酮和黄酮醇生物合成通路和次生代谢产物生物合成通路等方面。【结论】不同地区薜荔叶片类黄酮的含量和种类均较丰富,地区间类黄酮积累模式具有特异性;海南定安薜荔样本的类黄酮种类和含量最高,可作为重要的薜荔种质资源进行开发利用。【Objective】The species and content differences of flavonoids in leaves of Ficus pumila from different regions were studied to provide a reference for developing the evaluation standards of the quality of F.pumila,and for the conservation,development and utilization of germplasm resources.【Method】The leaf samples of F.pumila were collected from Wenchang of Hainan,Ding’an of Hainan,Guangzhou of Guangdong and Zhangzhou of Fujian.After crude extraction,the flavonoids from the leaves were qualitatively and quantitatively analyzed by using quantitative metabolomics technology.On this basis,the metabolic differences of flavonoids in F.pumila leaves from different regions were studied through PCA,heatmap clustering analysis,differential metabolite screening based on OPLS-DA and KEGG enrichment analysis.【Result】A total of 14 classes and 96 flavonoid compounds were identified,and the type of flavones and flavonols was large,each had 24 types of flavonoid compounds.F.pumila from Ding’an had the most abundant types and contents of flavonoids,respectively 77 types and 2.15μmol/g.The abundance of(-)-catechin was the highest in the four regions,which respectively were 1.45μmol/g(Ding’an),1.23μmol/g(Zhangzhou),1.20μmol/g(Guang�zhou)and 1.04μmol/g(Wenchang),respectively;Rutin,(-)-epicatechin and kaempferol 3-O-rutinoside also accounted for a large proportion of the total flavonoids,and some compounds were only enriched in specific regions.Cynaroside,for example,was detected only in samples from Wenchang.The flavonoid accumulation patterns of samples from different regions were quite different and had regional specificity.The differential metabolites were mainly concentrated in metabolic pathway,flavone and flavonol biosynthesis pathway and biosynthesis pathway of secondary metabolites.【Conclusion】The content and species of flavonoids in F.pumila leaves from different regions are abundant,and the accumulation pattern of flavonoids in different regions is specific.The types and content of flavonoids in
