摘  要:采用正相和反相硅胶柱色谱、半制备型高效液相色谱和Sephadex LH-20凝胶柱色谱等分离技术,从蓼科药用植物牛皮消蓼(Fallopia cynanchoides(Hemsl.)Harald.var.cynanchoides)中分离得到17个化合物,并通过核磁共振、质谱等多种波谱学技术鉴定化合物的化学结构,分别为lyoniresinol(1)、β-D-(1-O-acetyl-3,6-O-trans-diferuloyl)-fructofuranosyl-α-D-2′,4′,6′-O-triacetylglucopyranoside(2)、反式阿魏酸对羟基苯乙酯(3)、(3S,5R,6S,7E)-3,5,6-trihydroxy-7-megastigmen-9-one(4)、2,3-二羟基-1-(4-羟基-3,5-二甲氧基苯基)-1-丙酮(5)、2-methyl-5-(2′-oxo-4′-hydroxypentyl)-7-hydroxychromone(6)、2-(2′-hydroxypropyl)-5-methyl-7-hydroxychromone(7)、2,5-dimethyl-7-hydroxychromone(8)、2-methyl-5-acetonyl-7-hydroxychromone(9)、大黄素(10)、槲皮素(11)、(+)-花旗松素(12)、3,7-二甲氧基-5,3′,4′-三羟基黄酮(13)、咖啡酸甲酯(14)、原儿茶酸乙酯(15)、(2aS,3aS)-lyciumamide D(16)和3-acetyl-4,5-dimethoxy-benzoic acid(17)。化合物1~7、9、13、14、16、17为首次从何首乌属植物中分离得到,化合物1~9、11~17为首次从牛皮消蓼中分离得到。体外抗氧化活性评价结果显示,牛皮消蓼的70%乙醇提取物具有一定的ABTS+自由基清除能力,IC_(50)为14.21μg/mL;化合物1~3、11、13~17具有一定的ABTS+自由基清除能力,其中化合物1、13、14的IC_(50)优于阳性对照水溶性维生素E(IC_(50)=23.55μmol/L),分别为15.30、16.97和17.53μmol/L。本研究为药用植物牛皮消蓼的开发与利用提供了科学依据。To study the chemical constituents of the whole plant of Fallopia cynanchoides(Hemsl.)Harald.var.cynanchoides,seventeen compounds were isolated and purified by various modern chromatographic techniques,including normal phase and reverse phase silica gel column chromatography,Sephadex LH-20 column chromatography,semi-preparative HPLC.NMR,MS and other related spectroscopic analyses were used to identify their structures.The isolated compounds were identified as lyoniresinol(1),β-D-(1-O-acetyl-3,6-O-trans-diferuloyl)-fructofuranosyl-α-D-2′,4′,6′-O-triacetylglucopyranoside(2),p-hydroxyphenethyl trans-ferulate(3),(3 S,5 R,6 S,7 E)-3,5,6-trihydroxy-7-megastigmen-9-one(4),2,3-dihydroxy-1-(4-hydroxy-3,5-dimethoxyphenyl)-1-propanone(5),2-methyl-5-(2′-oxo-4′-hydroxypentyl)-7-hydroxychromone(6),2-(2′-hydroxypropyl)-5-methyl-7-hydroxychromone(7),2,5-dimethyl-7-hydroxychromone(8),2-methyl-5-acetonyl-7-hydroxychromone(9),emodin(10),quercetin(11),(+)-taxifolin(12),3,7-dimethoxy-5,3′,4′-trihydroxy-flavone(13),caffeic acid methyl ester(14),protocatechuic acid ethyl ester(15),(2a S,3a S)-lyciumamide D(16),and 3-acetyl-4,5-dimethoxy-benzoic acid(17).This was the first report of the presernce of compounds 1-7,9,13,14,16,and 17 in the genus Fallopia.Compounds 1-9 and 11-17 were first identified from F.cynanchoides var.cynanchoides.The 70%ethanol extract showed obvious ABTS+radicals scavenging effect with IC_(50)value of 14.21μg/mL;further in vitro bioassay indicated that compounds 1-3,11,13-17 showed moderate ABTS+radicals scavenging activities.Among them,compounds 1,13,14 demonstrated significant antioxidant effects with IC_(50)values of 15.30,16.97 and 17.53μmol/L,which were better than that of positive medicine trolox(IC_(50)23.55μmol/L).These findings provided an important foundation for the development and utilization of plant resource of F.cynanchoides var.cynanchoides.
