摘　　要：目的探究Ⅰ型胶原α1(collagen typeⅠalpha 1,COL1A1)对肺腺癌(lung adenocarcinoma,LUAD)恶性进程的影响。方法TIMER数据库用于分析泛癌中COL1A1基因表达情况;UCSC Xena数据库用于分析LUAD中COL1A1基因表达情况;Kaplan-Meier用于分析COL1A1表达与LUAD患者总生存率和无进展生存率的关系;定量逆转录聚合酶链式反应(quantitative reverse transcription PCR,qRT-PCR)用于检测LUAD组织和细胞中COL1A1 mRNA水平;CCK-8法、克隆形成实验、TUNEL实验、划痕实验和Transwell侵袭实验分别用于检测LUAD细胞的增殖、凋亡、迁移和侵袭;Western Blot用于检测COL1A1的蛋白表达。结果COL1A1在16种癌组织中表达上调。COL1A1在LUAD患者的肿瘤样本中高表达(P<0.001),COL1A1高表达与LUAD患者总生存率和无进展生存率较差显著相关(P<0.001)。与si-NC比较,COL1A1敲除在体外抑制了LUAD细胞的增殖、迁移和侵袭,并促进细胞凋亡(P<0.01)。结论COL1A1在LUAD中高表达,并与肿瘤进展和不良预后呈正相关,COL1A1可能是治疗LUAD新的预后生物标志物。Objective To explore the effect collagen type I alpha 1(COL1A1)on the malignant progression of lung adenocarcinoma(LUAD)cells.Methods The expression of COL1A1 gene in pan-cancer was analyzed by TIMER database.The expression of COL1A1 gene in LUAD was analyzed by UCSC Xena database.The relationship between COL1A1 expression and the overall survival and progression-free survival of LUAD patients was analyzed by Kaplan-Meier.The expression of COL1A1 in LUAD tissues and cells were detected using quantitative reverse transcription polymerase chain reaction(qRT-PCR).The proliferation,apoptosis,migration and invasion of LUAD cells were detected by CCK-8,colony formation,TUNEL,wound healing and Transwell invasion assay,respectively.The expression of COL1A1 protein was detected by Western blot.Conclusion The expression of COL1A1 was up-regulated in 16 cancer tissues.COL1A1 was highly expressed in tumor samples of LUAD patients(P<0.001),and the high expression of COL1A1 was significantly correlated with overall survival and progression free survival in LUAD patients(P<0.001).Compared with si-NC,COL1A1 knockout inhibited the proliferation,migration,and invasion of LUAD cells in vitro,and promoted cell apoptosis(P<0.01).Conclusion COL1A1 was highly expressed in LUAD and positively correlated with tumor progression and poor prognosis.COL1A1 might be a new prognostic biomarker for the treatment of LUAD.
