摘  要:旨在以大鼠为模型,研究竹叶黄酮(Bamboo leaf flavonoids,BLF)对敌草快(Diquat,DQ)应激动物肝脏损伤、抗氧化功能及相关基因表达的影响。选取32只5周龄雄性Sprague-Dawley(SD)大鼠,将其随机分为4组:对照组(Con)、1000mg/kg竹叶黄酮添加组(BLF)、DQ应激组(DQ)、1000mg/kg竹叶黄酮+DQ应激组(BLF-DQ)。结果显示,与Con组相比,DQ组大鼠血清谷草转氨酶(AST)的含量显著降低(P<0.05),肝脏总抗氧化能力(T-AOC)、谷胱甘肽过氧化物酶(GPX)、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)水平显著减少(P<0.05),丙二醛(MDA)含量显著升高(P<0.05)。同时,肝脏血红素氧合酶-1(HO-1)、GPX、CAT、SOD1和核因子E2相关因子(Nrf2)的基因表达量显著降低(P<0.05);灌胃1000mg/kg BLF组大鼠血清AST和谷丙转氨酶(ALT)含量显著降低(P<0.05),肝脏T-AOC、GPX、CAT和SOD水平显著增强(P<0.05),肝脏HO-1、GPX、CAT、SOD1、Nrf2和醌氧化还原酶1(NQO1)基因表达水平显著上调(P<0.05);与DQ组相比,灌胃1000mg/kg BLF大鼠肝脏指数显著降低(P<0.05),血清中AST和ALT含量显著减少(P<0.05),同时肝脏中CAT、GPX、T-AOC水平显著增加(P<0.05),MDA含量显著降低(P<0.05),肝脏HO-1、GPX、CAT、SOD1和Nrf2的基因表达量显著上调(P<0.05)。结果表明,BLF能够缓解DQ应激引起的大鼠肝脏损伤,改善肝脏抗氧化功能抑制状态,激活Nrf2信号通路和PINK/Parkin线粒体自噬相关基因表达。Bamboo leaf flavonoids(BLF)are compounds extracted from bamboo leaves,possessing properties including antioxidant,antimicrobial and anti-inflammatory properties.This study aimed to investigate the effects of BLF on liver damage,antioxidant function,and related gene expression in rats induced by diquat(DQ).Thirty-two 5-week-old male Sprague-Dawley(SD)rats were randomly divided into four experimental groups:the control group(Con),1000mg/kg BLF group(BLF),DQ stress group(DQ),and 1000mg/kg BLF+DQ stress group(BLF-DQ).The results showed that compared to the Con,the DQ group exhibited significantly decreased serum AST levels(P<0.05),as well as decreased levels of T-AOC,GPX,SOD,and CAT in the liver(P<0.05),and increased MDA levels in rats(P<0.05).Additionally,the gene expression levels of HO-1,GPX,CAT,SOD1,and Nrf2in the liver were significantly reduced(P<0.05).In contrast,1000 mg/kg BLF significantly decreased serum AST and ALT levels(P<0.05),increased levels of TAOC,GPX,CAT,and SOD in liver(P<0.05),and significantly increased gene expression of HO-1,GPX,CAT,SOD1,Nrf2,and NQO1(P<0.05).Compared to the DQ group,BLF-DQ significantly decreased liver index(P<0.05),reduced serum AST and ALT levels(P<0.05),increased levels of CAT,GPX,and T-AOC in liver(P<0.05),decreased MDA levels(P<0.05),and significantly upregulated gene expression levels of HO-1,GPX,CAT,SOD1,and Nrf2(P<0.05).These findings indicated that BLF alleviate liver damage caused by DQ stress in rats,improve liver antioxidant function inhibition,activate the Nrf2signaling pathway and PINK/Parkin mitophagy-related gene expression.
