摘　　要：目的观察2,3-丁二酮单肟钾盐的抑菌和病毒杀灭效果,并检测其皮肤刺激性,为酮肟钾盐的实际应用提供依据。方法采用肉汤稀释法、透射电子显微镜观察、间接免疫荧光染色检测和动物试验方法,对2,3-丁二酮单肟钾盐的抑菌和病毒灭活效果及皮肤刺激性进行观察。结果2,3-丁二酮单肟钾盐对金黄色葡萄球菌、大肠埃希菌和鼠伤寒沙门菌的最小抑菌浓度均为12.5 mg/mL;以浓度100 mg/mL作用10 min,对3种试验指示菌抑菌率均>99.9%;透射电子显微镜观察显示,该酮肟钾盐对大肠埃希菌细胞膜具有明显损伤作用。该酮肟钾盐与乙型脑炎病毒或肠道病毒71型孵育后,间接免疫荧光法检测细胞中病毒感染为阴性;试验浓度下,该酮肟钾盐对白色豚鼠皮肤刺激性小。结论2,3-丁二酮单肟钾盐对细菌和病毒具有较强的杀灭效果,对实验动物完整皮肤的刺激性较轻。Objective To observe the bacteriostatic and virucidal effects of potassium salt of 2,3-butanedione monoxime and test its skin irritation,so as to provide a basis for the practical application of potassium salt of ketoxime.Methods Broth dilution method,transmission electron microscopy observation,indirect immunofluorescence staining assay and animal testing methods were used to observe the bacteriostatic and viral inactivation effects and skin irritation of potassium salt of 2,3-butanedione monoxime.Results The minimum inhibitory concentration of 2,3-butanedione monoxime potassium salt on Staphylococcus aureus,Escherichia coli and Salmonella typhimurium was 12.5 mg/mL.The inhibition rate of the three test indicator bacteria was>99.9%after 10 min of action at a concentration of 100 mg/mL,and the damage of the cell membranes of Escherichia coli were obvious by the ketoxime potassium salt under the transmission electron microscopy observation.Encephalitis B virus or enterovirus 71 was negative for viral infection in cells by indirect immunofluorescence method after incubation with the ketoxime potassium salt.The ketoxime potassium salt had little irritation to the skin of white guinea pigs under the test concentration.Conclusion Potassium salt of 2,3-butanedione monoxime has strong killing effect on bacteria and viral and mild irritant to the intact skin of laboratory animals.
