摘  要:目的探讨甘草酸通对脑缺血再灌注(I/R)损伤的神经保护作用及其分子机制。方法采用胎鼠大脑皮质神经元氧糖剥夺(OGD)模型和大鼠大脑中动脉闭塞(MCAO)模型,分别在体外和体内模拟脑I/R损伤。结果本研究证实甘草酸对神经元增殖有促进作用,对神经元焦亡有抑制作用。此外,发现甘草酸可以改善脑梗死。Western blot结果显示,甘草酸下调OGD和MCAO模型中核苷酸结合寡聚结构域、富含亮氨酸重复序列和含1个Pyrin结构域(NLRP1)、半胱氨酸天冬氨酸特异性蛋白酶1(caspase-1)、gasdermin D(GSDMD)、IL-1β、IL-6、TNF-α和iNOS蛋白的表达。结论甘草酸可通过抑制NLRP1依赖性神经元焦亡,对脑I/R损伤具有神经保护作用。Objective To investigate the neuroprotective effect of glycyrrhizin on cerebral ischemia reperfusion(I/R)injury and its molecular mechanism.Methods This study used a fetal rat cortical neuron oxygen glucose deprivation(OGD)model and a rat middle cerebral artery occlusion(MCAO)model to simulate cerebral I/R injury in vitro and in vivo,respectively.Results This study confirmed that glycyrrhizic acid can promote neuronal proliferation and inhibit neuronal pyroptosis.In addition,glycyrrhizin was found to improve cerebral infarction.Western blot results showed that glycyrrhizin down-regulated the expression of nucleotide-binding oligomerization domain,leucine-rich repeat and pyrin domain-containing 1(NLRP1),cysteinyl aspartate-specific protease 1(caspase-1),gasdermin D(GSDMD),IL-1β,IL-6,TNF-αand iNOS proteins in OGD and MCAO models.Conclusion Glycyrrhizic acid has neuroprotective effect on cerebral ischemia reperfusion I/R injury by inhibiting NLRP1-dependent neuronal pyroptosis.
