摘 要:为研究不同低温处理对青花菜黄化衰老过程中叶绿素变化及叶绿素降解相关基因表达量的影响。以兰州高原特色蔬菜‘耐寒优秀’品种的青花菜为试验材料,设置近冰温(-0.7℃~-0.4℃)、4℃、10℃3个贮藏温度。结果表明,近冰温(-0.7℃~-0.4℃)处理组在整个贮藏期叶绿素含量显著高于4℃、10℃处理,且L^(*)、a^(*)和b^(*)显著低于4℃和10℃处理。在整个贮藏期,近冰温处理的青花菜BoCLH1(叶绿素酶1)和BoCLH2(叶绿素酶2)的表达量显著低于4℃、10℃处理,这说明近冰温处理抑制了青花菜贮藏期BoCLH1(叶绿素酶1)和BoCLH2(叶绿素酶2)的表达,而4℃、10℃处理诱导了青花菜贮藏期BoCLH1(叶绿素酶1)和BoCLH2(叶绿素酶2)的表达;近冰温处理的青花菜BoPPH(脱镁叶绿素酶)的表达量随着贮藏时间的延长不断下降,而在4℃和10℃处理组中在贮藏15 d时有显著上升,且始终高于近冰温处理组。近冰温处理的BoPaO(脱镁叶绿酸a加氧酶)表达量高峰出现明显滞后,而BoRCCR(红色叶绿素代谢产物还原酶)的表达量在贮藏过程中始终显著高于4℃和10℃处理组。可见,近冰温处理可有效抑制叶绿素的降解,延缓青花菜组织衰老。This study investigates effects of different low temperature treatments on the changes in chlorophyll content and the expression of chlorophyll degradation-related genes during the yellowing and senescence of broccoli,with"cold-tolerant excellent"variety from Lanzhou plateau summer vegetable as the experimental material.Three storage temperatures were set:slightly frozen(-0.7℃--0.4℃),4℃,and 10℃.The results indicated that the content of chlorophyll under the sub-zero(-0.7℃--0.4℃)treatment group was significantly higher compared to the 4℃ and 10℃ treatments throughout the storage period,while the values of L^(*),a^(*),and b^(*)were significantly lower in the sub-zero treatment.The expression levels of BoCLH1(chlorophyllase 1)and BoCLH2(chlorophyllase 2)in broccoli treated with sub-ice temperature were significantly lower than those treated with 4℃ and 10℃.The sub-freezing temperature treatment inhibited the expression of BoCLH1(chlorophyllase 1)and BoCLH2(chlorophyllase 2)during storage in broccoli,while treatments at 4℃ and 10℃ induced the expression of BoCLH1(chlorophyllase 1)and BoCLH2(chlorophyllase 2)during storage in broccoli;as the storage time extended,the expression level of Boophone Phenolase(BoPPH)in broccoli decreased steadily after near-ice temperature treatment,while it significantly increased in the 4℃ and 10℃ treatment groups after 15 days of storage,and remained higher than that in the near-ice temperature treatment group throughout.The expression peak of BoPaO(Pheophytin a oxygenase)was significantly delayed after the near-freezing treatment,and the expression of BoRCCR(Red chlorophyll metabolite reductase)remained significantly higher throughout the storage period compared to the 4℃ and 10℃ treatment groups.In conclusion,near-freezing temperature treatment effectively inhibits the degradation of chlorophyll and delays the senescence in brassica tissues.
