摘  要:非典型趋化因子受体(atypical chemokine receptors,ACKRs)指的是一类不能通过经典G蛋白进行信号转导的、具有七次跨膜结构域(TM1~TM7)的蛋白,分布于多种细胞、组织和器官;能结合多种配体,还能与经典趋化因子受体(conventional chemokine receptors,c CKRs)形成异源二聚体。ACKRs通过调节趋化因子的丰度、定位及可用性发挥着多种功能,包括趋化因子内吞、转胞吞、清除、提呈和贮存。ACKRs对正常人体的生物学功能的发挥及各种病理过程的发生发展均起着重要作用。本文就ACKRs亚家族的每个成员及其配体特征、功能及相互作用机制进行综述,以期为将来开发有效预防和治疗人体多种疾病的靶向药物提供参考。Atypical chemokine receptors(ACKRs)are seven-transmembrane domain-contained proteins that are unable to transduce signals through classical G-protein pathways.They are distributed throughout various cells,tissues,and organs,and are capable of binding to multiple ligands.Additionally,they can form heterodimers with cCKRs.ACKRs play various roles in modulating the abundance,localization,and availability of chemokines.The functions of ACKRs include chemokine endocytosis,transcytosis,scavenging,presentation,and reservoir.ACKRs play crucial roles in normal biological functions and the occurrence and development of various pathological conditions in human body.This review summarizes the characteristics,functions,and interaction mechanisms of each member of the ACKRs subfamily and their respective ligands.It provides a reference for the future development of targeted drugs for the effective prevention and treatment of various human diseases.
