摘  要:目的:探究长链非编码RNA00941(Linc00941)、微小RNA-145(miR-145)表达及转化生长因子β1/Smad(TGF-β1/Smad)通路相关因子在胃癌组织中的表达与意义。方法:选取收治的51例胃癌患者,手术治疗后经病理学检查确诊。收集其胃癌组织及其癌旁组织,比较其中Linc00941、miR-145及TGF-β1/Smad通路相关因子表达差异;提取患者的胃癌组织细胞进行组织培养,并对其进行Linc00941、miR-145基因的转染、沉默,分析其对胃癌细胞的增殖、迁移和侵袭能力的影响。结果:与癌旁组织比较较,胃癌组织患者的Linc00941及其信使核糖核酸(mRNA)相对表达水平升高,miR-145及其mRNA相对表达水平降低(均P<0.05)。胃癌组织中的TGF-β1、转化生长因子β受体Ⅱ(TGF-βRⅡ)及Smad2/3均较癌旁组织显著降低(均P<0.05)。NC组、Linc00941转染组及Linc00941沉默组三组间吸光值(A值)及凋亡率两两比较均存在统计学差异(均P<0.05),Linc00941沉默组在培养24、48、72 h时的A值低于NC组,凋亡率高于NC组;Linc00941转染组各时间的A值高于NC组,凋亡率低于NC组(均P<0.05)。与NC组比较较,Linc00941沉默组的细胞迁移数目及细胞侵袭数目均降低,Linc00941转染组均升高,组间两两比较差异存在统计学意义(均P<0.05)。与NC组比较较,miR-145转染组培养24、48、72 h的A值均降低,凋亡率升高;miR-145沉默组的A值升高,细胞凋亡率降低(均P<0.05)。与NC组比较较,miR-145转染组的细胞迁移数目及细胞侵袭数目均降低,miR-145沉默组均升高,组间两两比较差异存在统计学意义(均P<0.05)。结论:在胃癌组织中,Linc00941及其mRNA相对表达水平升高,miR-145及其mRNA相对表达水平降低,TGF-β1/Smad通路相关因子TGF-β1、TGF-βRⅡ及Smad2/3表达降低,且Linc00941、miR-145表达影响胃癌细胞的增殖、侵袭和迁移。Objective:To investigate the expression and significance of Linc00941,miR-145 and TGF-β1/Smad pathway related factors in gastric cancer tissues.Methods:A total of 51 cases of gastric cancer were selected,and the diagnosis was diagnosed after surgery.Gastric cancer tissues and adjacent tissues of patients were collected,and the expression differences of Linc00941,miR-145 and TGF-β1/Smad pathway related factors were compared.Gastric cancer tissue cells were extracted from patients for tissue culture,and transfected and silenced Linc00941 and miR-145 genes to analyze their effects on the proliferation,migration and invasion of gastric cancer cells.Results:Compared with paracancer tissues,the relative expression levels of Linc00941 and its mRNA were increased in patients with gastric cancer tissues,while the relative expression levels of miR-145 and its mRNA were decreased(all P<0.05).The levels of TGF-β1,transforming growth factorβreceptorⅡ(TGF-βR)and Smad2/3 in gastric cancer tissues were significantly lower than those in adjacent tissues(all P<0.05).There were statistically significant differences in absorbance value(A value)and apoptosis rate among NC group,Linc00941 transfection group and Linc00941 silencing group(all P<0.05).The A value of Linc00941 silenced group were lower than that of NC group at 24,48 and 72 hours,and the apoptosis rate was higher than that of NC group(all P<0.05).The A value of Linc00941 transfection group was higher than that of NC group,and the apoptosis rate was lower than that of NC group(all P<0.05).Compared with NC group,the number of cell migration and cell invasion decreased in Linc00941 silencing group,and increased in Linc00941 transfection group,and the pairings between groups were statistically significant(all P<0.05).Compared with the NC group,the A value of miR-145 transfected group was decreased for 24,48 and 72 hours,and the apoptosis rate was increased(all P<0.05);in the miR-145 silencing group,the A value was increased,and the apoptosis rate was decreased(all P<0.0
