摘  要:目的应用去势法构建肾阳虚型肌少-骨质疏松症(OS)大鼠模型,并从肌骨能量代谢角度进行机制探讨。方法60只3月龄雌性SD大鼠随机分为去势1、2、3组与对照1、2、3组,共6组,分别在术后4、8、12周依次处死取材。去势组行双侧卵巢切除术,对照组行假手术,饲养期间观察动物行为,测体质量、抓力,采集血清检测血清碱性酸酶(ALP)、抗酒石酸酸性磷酸酶(TRAP)、雌二醇(E_(2))、鸢尾素(Irisin),采集左后肢三头肌、左股骨行形态学检测、免疫组化观察鸢尾素表达情况,称量右股四头肌湿重,右股骨行骨密度检测,之后行蛋白免疫印迹分析(WB)检测观察鸢尾素表达情况。结果术后4周,去势组大鼠抓力明显下降(P<0.05),免疫组化结果显示去势组骨骼肌及骨组织中鸢尾素表达明显下降(P<0.05),但组间WB差异无统计学意义。术后第6周,去势组开始出现肾阳虚证表征。术后8周,两组间骨组织鸢尾素表达无明显差异(P>0.05),血清TRAP含量明显增加(P>0.05),余指标均明显下降,形态学变化符合趋势。术后12周,去势组只有血清ALP含量显著高于对照组(P<0.01),其余指标变化趋势与4周前相同。结论去势法可成功构建肾阳虚型肌少-骨质疏松症模型。模型动物骨骼肌鸢尾素表达下降与该病发生发展呈正相关,肌骨间能量代谢下降可能是该病致病机制之一。Objective Castration method was used to construct a rat model of kidney-Yang deficiency with sarcopenia-osteoporosis and explore the correlation between its pathogenesis and musculoskeletal energy metabolism.Methods Sixty 3-month-old female SD rats were randomly divided into castration groups 1,2 and 3 and control groups 1,2 and 3,and then killed at 4,8 and 12 weeks after operation,respectively.The castration groups underwent bilateral oophorectomy,and the control groups underwent sham operation.During feeding,the animal behavior was observed and the body weight and grip strength were measured.The serum was collected to detect the levels of alkaline phosphatase(ALP),tartrate resistant acid phosphatase(TRAP),estradiol(E_(2))and irisin.The left hind limb triceps and left femur were collected for morphological detection and immunohistochemistry was used to observe irisin expression.The right quadriceps wet weight was weighed and the right femur bone density was detected.Western Blot detection was performed to observe irisin expression.Results At 4 weeks after operation,the gripping power of rats in the castration groups was significantly decreased(P<0.05),and the expression of irisin in skeletal muscle and bone tissue of castration groups was significantly decreased by immunohistochemistry(P<0.05),but there was no significant difference in Western Blot among groups.At the 6th week after operation,kidney-Yang deficiency began to appear in the castration groups.At 8 weeks after operation,there was no significant difference in irisin expression in bone tissue between castration group and control group(P>0.05).The serum TRAP content was significantly increased(P>0.05),and the remaining indexes were significantly decreased,and the morphological changes were in line with the trend.At 12 weeks after operation,only serum ALP content in the castration groups was significantly higher than that in the control groups(P<0.01),and the change trend of other indexes was the same as that 4 weeks ago.Conclusion Castration method can
