摘 要:目的优化反胶束法萃取水飞蓟蛋白工艺,探究该方法对水飞蓟蛋白理化性质的影响。方法通过单因素试验和响应面试验优化反胶束法萃取水飞蓟蛋白工艺,分析提取因素对蛋白提取率的影响;对比碱提酸沉法,分析反胶束法水飞蓟蛋白溶解度、持油性、起泡性、起泡稳定性、乳化性及乳化稳定性。结果反胶束法萃取水飞蓟蛋白最佳提取工艺参数为含水量20、液固比201(mL g)、前萃取pH 8.0、前萃取温度40℃、后萃取时间90 m in、后萃取pH 8.0,该条件下水飞蓟蛋白提取率为(60.44±0.20)%;反胶束法萃取的水飞蓟蛋白溶解度、持油性、起泡性、乳化性和乳化稳定性均高于碱提酸沉法提取的水飞蓟蛋白。结论反胶束法提取水飞蓟蛋白较传统方法更具优势,在水飞蓟蛋白开发中具有较好的应用前景。Objective To optimize the extraction process of Silybum marianum protein by the reverse micelle met-hod and explore the effect of this method on the physicochemical properties of S.marianum protein.Method The effect of the extraction factor on protein yield was investigated by using single factor experiments and response surface optimization.The reverse micelle method on solubility,oil retention,foaming capacity,foaming stability,emulsification activity,and emulsification stability of S.marianum protein was analyzed by comparing alkali extraction and acid precipitation method.Results The optimal extraction parameters of S.marianum protein by the reverse micelle method were as follows:the water content was 20,the liquid-solid ratio was 201(mL g),the pH of forward extraction was 8.0,the temperature of forward extraction was 40℃,the time of backward extraction was 90 min,and the pH of backward extraction was 8.0.Under these conditions,the extraction rate of S.marianum protein was(60.44±0.20)%.The solubility,oil retention,foaming capacity,emulsification activity,and emulsification stability of S.marianum protein extracted by the reverse micelle method were higher than those extracted by alkali extraction and acid precipitation method.Conclusion The reverse micelle method for the extraction of S.marianum protein has advantages over the conventional method and is promising for the develop-ment of S.marianum protein.
