摘 要:目的探讨姜黄素对人雄激素非依赖性前列腺癌C4-2细胞和典型人雄激素依赖性前列腺癌LNCaP细胞增殖、侵袭及细胞周期的影响。方法取对数生长期C4-2细胞和LNCaP细胞,分为正常对照组,20、40、60、80μmol/L姜黄素组。姜黄素处理24 h后,采用MTT法检测各组细胞增殖活性;选取20、40μmol/L姜黄素组作为试验组,未处理组作为对照组,各组细胞处理24 h后,采用流式细胞术检测经姜黄素处理的C4-2细胞和LNCaP细胞的细胞周期;采用Transwell小室试验检测对照组C4-2、LNCaP细胞及20、40μmol/L姜黄素组细胞体外侵袭数量。结果MTT结果显示,与正常对照组比较,不同浓度姜黄素组C4-2细胞和LNCaP细胞增殖活性明显降低(P<0.05或P<0.01);流式细胞术检测结果显示,应用20、40μmol/L姜黄素处理后,G_(2)/M期阻滞且G_(2)/M期细胞比例均高于正常对照组(P<0.05或P<0.01);Transwell小室试验结果显示,与正常对照组比较,姜黄素处理组C4-2细胞和LNCaP细胞体外侵袭数量明显降低(P<0.01)。结论姜黄素对人前列腺癌C4-2细胞、LNCaP细胞的增殖、侵袭具有抑制作用,且使细胞周期阻滞在G_(2)/M期。Objective To investigate the effects of curcumin on the proliferation,invasion and cell cycle of C4-2 cells of human androgen-independent prostate cancer and LNCaP cells of typical human androgen-dependent prostate cancer.Method C4-2 cells and LNCaP cells at logarithmic growth stage were divided into normal control group and 20,40,60 and 80μmol/L curcumin groups.After treated curcumin for 24 h,the cell prolife-ration activity of each group was detected by MTT assay.20 and 40μmol/L curcumin groups were selected as the experimental group,and the untreated group as the control group.After 24 h treatment,the cell cycle changes of C4-2 cells and LNCaP cells treated with curcumin were detected by flow cytometry.The invasion ability of C4-2 and LNCaP cells in control group and 20,40μmol/L curcumin group in vitro was detected by Transwell assay.Results MTT results showed that,the proliferative activity of C4-2 cells and LNCaP cells in curcumin groups with different concentrations decreased significantly compared with normal control group(P<0.05 or P<0.01).Flow cytometry showed that G_(2)/M phase was blocked and the proportion of G_(2)/M phase cells was higher than that of normal control group after treatment with 20 and 40μmol/L curcumin(P<0.05 or P<0.01).The results of Transwell laboratory experiment showed that the invasion ability of C4-2 cells and LNCaP cells in curcumin treated group was significantly decreased comparing with the normal control group(P<0.01).Conclusion Curcumin can inhibit the proliferation and invasion of human prostate cancer C4-2 cells and LNCaP cells,and arrest the cell cycle in G_(2)/M phase.
