摘  要:目的探讨组蛋白的甲基转移酶2抑制剂BAY-598和阿霉素在骨肉瘤中的协同抗癌作用。方法通过细胞的增殖、迁移、侵袭和集落形成实验,评估两种药物单独或联合使用后对骨肉瘤细胞恶性特征的抑制作用。采用流式细胞术检测细胞凋亡和细胞周期情况。结果组蛋白甲基转移酶2抑制剂和阿霉素联合治疗可协同抑制细胞的生长、迁移、侵袭,诱导细胞凋亡,导致OS细胞细胞周期阻滞。进一步的机制探索表明,JAK-STAT信号通路可能是协同作用的关键。结论组蛋白甲基转移酶2抑制剂和阿霉素对骨肉瘤具有协同抗癌作用,它们的联合使用可能是一种很有前途的骨肉瘤治疗策略。Objective To investigate the synergistic anticancer effect of BAY-598,a histone methyltransferase 2 inhibitor,and doxorubicin in osteosarcoma.Methods The inhibitory effects of the 2 drugs,either alone or in combination,on the malignant characteristics of osteosarcoma cells were assessed through experiments involving cell proliferation,migration,invasion,and colony formation.Flow cytometry was employed to detect cell apoptosis and the cell cycle.Results Combined treatment with histone methyltransferase 2 inhibitors and doxorubicin synergistically inhibited cell growth,migration,and invasion,inducing cell apoptosis and leading to cell cycle arrest in osteosarcoma cells.Further mechanistic exploration suggested that the JAK-STAT signaling pathway might be a key player in this synergistic effect.Conclusion Histone methyltransferase 2 inhibitors and doxorubicin have synergistic anticancer effects on osteosarcoma,and their combined use may be a promising treatment strategy for osteosarcoma.
